Auranofin, gambogic acid (GA), disulfiram, menadione, oltipraz, parthenolide, plumbagin from Plumbago indica, PZQ, thonzonium bromide, sanguinarine chloride hydrate, dimethyl sulphoxide (DMSO), percoll and fetal bovine serum (FBS) were from Sigma-Aldrich. The Ro 15–5458 compound was a kind gift from Dr H. Stohler (Hoffman-La Roche, Basel, Switzerland) and oxamniquine was provided by Pfizer, London. Drugs were dissolved in DMSO to obtain stock solutions at 10 mM and were then diluted into culture medium. CellTiter-Glo (CTG) reagent, used in the schistosomula viability luminescence-based assay, and CellTox green dye, used in the schistosomula staining, were from Promega. BioWhittaker Dulbecco-Modified Eagle’s Medium (DMEM) lacking phenol red and containing 4500 mg/l glucose, 1 mM Hepes pH 6.98–7.30, 2 mM L-glutamine, 1x antibiotic-antimycotic reagent (Life Technologies) and 10% heat inactivated FBS, was used as tissue culture medium for schistosomula. Adult worms were cultured in BioWhittaker DMEM containing 4500 mg/l glucose, 2 mM L-glutamine, 100 U/ml penicillin, 100 μg/ml streptomycin, 0.5 μg/ml amphothericin B and 10% heat inactivated FBS.

Cell viability was evaluated using CCK8 (Beyotime, Jiangsu, China). Cells (2 × 103) were seeded in 96-well plates by different treatment, and cultured for different time points. CCK8 solution (10 μl) was added to each well, and the plates were incubated at 37°C for 1 hour. Absorbance was measured at 450 nm on a microplate reader (Bioteck). DMEM containing 10% CCK8 was used as a control. Proliferating cell count was measured using the Cell-Light EdU DNA Cell Proliferation Kit (RIBOBio Co, Guangzhou, China). The cells were seeded in 96-well culture plates and exposed to media with or without plumbagin. 2000 cells/well were treated with 50 μmol/L of EdU for 2 h at 37 °C. After being fixed with 4% paraformaldehyde for 15 min, the cells were baptized with 0.5% Triton X-100 for 30 minutes and rinsed with PBS three times. Thereafter, the cells were exposed to 100 μL/well of 1×Apollo® reaction cocktail for 30 minutes and incubated with 5 μg/mL of Hoechst 33342 to stain the cell nuclei for 30 minutes. Images were captured using a fluorescent microscope (Olympus, Tokyo, Japan). Experiments were repeated at least three times.

Cellular tyrosinase activity was determined using a previously described method with modification [25 (link),26 (link)]. Cultured B16F10 cells were incubated with α-MSH in the absence or presence of plumbagin, and cells were then washed and lysed with PBS containing 1% sodium deoxycholate and 0.5% Triton X-100. After the protein concentration was determined, 50 μg of protein was incubated with 2.0 mM l-DOPA and 0.1 M PBS (pH 6.8) for 1 h at 37 °C. The oxidation of l-DOPA was measured at 475 nm (OD475) using an absorbance reader. Activity was measured using the following formula: tyrosinase activity (%) = (OD475 of sample/OD475 of control) × 100.

The construction of StDHOase [61 (link)], KpDHOase [62 (link)], huDHOase [61 (link)], and ScDHOase [61 (link),63 (link)] expression plasmids were reported previously. The recombinant protein was purified using the protocol described previously for SSB-like proteins [64 (link),65 (link),66 (link),67 (link),68 (link)]. Briefly, E. coli BL21(DE3) cells were transformed with the expression vector, and the overexpression of the expression plasmids was induced by incubating with 1 mM isopropyl thiogalactopyranoside. The protein was purified from the soluble supernatant by using Ni2+-affinity chromatography (HiTrap HP; GE Healthcare Bio-Sciences), eluted with buffer A (20 mM Tris–HCl, 250 mM imidazole, and 0.5 M NaCl, pH 7.9), and dialyzed against a dialysis buffer (20 mM Tris–HCl and 0.1 M NaCl, pH 7.9). The protein purity remained at >97% as determined using SDS–PAGE (Mini-PROTEAN Tetra System; Bio-Rad, CA, USA).

Accurately weighed 25mg standard Plumbagin was dissolved in 25 ml of methanol to prepare 1mg/ml solution. Preparation of Test samples: Each 1 gm of sample was taken in conical flask along with 10 ml methanol and kept it overnight. Next day all samples are sonicated and centrifuged. Supernatant liquid is collected and stored in test tube for further use. Application of standard and sample: For spot application, Linomat V sample applicator (CAMAG, 4132 Muttenz, Switzerland) was used and samples solution applied to Silica Gel 60 F254 plate as sharp bands. After that this plate kept aside till the spots get dried. After that the mobile phase, Toluene: Formic acid (9.9:0.1) was poured into Twin trough chamber and chamber covered with lid for 45 minutes to allow saturation. Then lid was removed and plate was inserted into the chamber carefully. Hereafter the plate was allowed to develop until the solvent front had travelled at the distance of 70mm above the base of the plate. After this the plate was taken off from the chamber and dried by using blow dryer and then at 266nm wavelength, detection was done by using CAMAG TLC Scanner III & Win cats 4.02 software. This study was done at CRL (Central Research Laboratory), DMIHER ( Datta Meghe Institute of Higher Education & Research), Sawangi (M), Wardha, Maharashtra.

PLU was purchased from Aladdin Reagent Ltd. in Shanghai, China. NIT, CPR, TMP, and CHL were obtained from Sigma-Aldrich, while MEC was sourced from Shanghai Yuanye Biological Co., Ltd. in Shanghai, China. The stock solutions of PLU and antibiotics were prepared in water (MEC) or dimethyl sulfoxide (DMSO) (NIT, CPR, TMP, CHL, and PLU) due to the varying solubility of each compound. The solutions were then passed through 0.22 μm filters and stored in the dark at -20°C. Before being used in assays, the stocks were thawed and diluted to the desired concentrations.
