Drosophila Genetics and Fly Food Preparation

Flies were cultured on yeast/molasses-based standard fly food (recipe: 10L H2O, 138g agar, 220g molasses, 750g malt extract, 180 dry yeast, 800g corn flour, 100g soy flour, 62.5ml propionic acid, 20g Methyl 4-Hydroxybenzoate, and 72ml ethanol). For GeneSwitch experiments, 86 mg RU486 was additionally dissolved in the ethanol for 200 μM final concentration. Flies were maintained at 25°C with a 12h light/dark cycle and 65% humidity. All flies used in the experiments reported were females.
Fly lines HmlΔ::Gal4, LSP2::Gal4, Elav::Gal4 and W1118 were provided by the Bloomington Drosophila Stock Center. RNAi line MANFRNAiv12834 (UAS::MANFRNAi) was obtained from the Vienna Drosophila RNAi Center. These construct has no predicted off-targets. MANFRNAiv12834 was combined with a transgene driving Dicer overexpression (UAS::DICER2) to achieve efficient MANF knock-down19 (link).

Rearing and Genetic Manipulation of Cabbage White Butterfly

P. rapae larvae were collected primarily from cabbage fields in the experimental farmland of Zhejiang University, Hangzhou, China and the laboratory colony of P. rapae was reared at 25 ± 1°C and 70% relative humidity under a photoperiod of 14 hr light: 10 hr dark in a greenhouse as previously described by Zhang et al. (2005) (link).
The following fly stocks were from the Bloomington Drosophila Stock Center: HmlΔ-Gal4 (30139); UAS-5-HT1B RNAi (27634, 25833); UAS-5-HT2B RNAi (25874, 60488). The 5-HT1B control and 5-HT1BΔIII-Vflies were generous gifts from Leslie Vosshall (Gasque et al., 2013 (link)).

Drosophila Strains for Genetic Studies

These Drosophila strains were obtained from the Bloomington Stock Center: He-GAL4 (RRID:BDSC_8699); HmlΔ-GAL4 (RRID:BDSC_30141); Lz-GAL4; UAS-GFP (RRID:BDSC_6313); UAS-GFP (RRID:BDSC_6874); UAS-TRE-Red (RRID:BDSC_59012). Pxn-GAL4 (FBtp0022490) was kindly donated by Utpal Banerjee. All “wild-type” strains were relevant GAL4 driver outcrossed to either UAS-GFP or to w; P(w+).
