Detailed Small Molecule Preparation

Penfluridol, astemizole, and terfenadine were obtained from Sigma‐Aldrich® (Zwijndrecht, The Netherlands). Penfluridol (P3371) and terfenadine (T9652) were diluted in absolute EtOH (EMSURE®) to a stock solution of 50 mm. Likewise, astemizole (A2861), sertindole (S8072), chlorprothixene (C1671), chlorpromazine (C8138), clemastine (SML0445), and loratadine (L9664) were diluted in dimethyl sulfoxide (DMSO; Sigma‐Aldrich®) to a stock solution of 50 mm (Table S1). Serial dilutions were established by diluting these stock solutions in the cell‐specific medium of the various cell lines (Table S1).

Antiprotozoal Screening of Small Molecules

Clemastine, astemizole, oryzalin, pyrimethamine, and combretastatin A4 stocks were obtained from Sigma-Aldrich (St. Louis, MO, USA). All compounds were dissolved in tissue-culture-grade DMSO (Sigma). In some cases, we compared pretreated and repeatedly (continuously) treated tachyzoites. In all, we compared the following four treatments: (1) untreated vehicle controls; (2) treatment alone (tachyzoites are exposed to drug during the assay); and (3) Clemastine-combined pretreatment and mid-treatment (tachyzoites are exposed during the prior lytic cycle and during the experiment).

Monoclonal AQP4-IgG and Compound Screening

Purified human monoclonal recombinant AQP4-IgG rAb-53 (AQP4-IgG) was provided by Dr. Jeffrey Bennett (Univ. Colorado Denver) as described [45 (link)]. Control human IgG (control-IgG) was purchased from Pierce Biotechnology (Rockford, IL, USA). Human complement (HC) was purchased from Innovative Research (Novil, MI, USA). Test drugs included clobetasol, miconazole, benztropine, clemastine, fumarate, retinoic acid and citicolone (Sigma-Aldrich, St. Louis, MO, USA), enprofylline, olesoxime and quetiapine fumarate (Santa Cruz Biotechnology, Dallas, TX, USA), GC-1 and quercetin (Tocris Bioscience, Bristol, UK), fasudil (Tszchem, Lexington, MA, USA), and Y-27632 (BD Biosciences, San Jose, CA, USA); Triiodothyronine (T3, Calbiochem, Billerica, MA, USA) was used as positive control. Drugs were dissolved in 2.5 % DMSO + 2 % solutole in PBS. Unless otherwise specified all other chemicals and media were purchased from Sigma-Aldrich.

Screening novel compounds against N. fowleri

The COVID Box, provided by Medicines for Malaria Venture (MMV, Geneva, Switzerland), is a collection of 160 commercially available drugs with known or predicted activity against the newly discovered pandemic virus, SARS-CoV-2, as shown in Supplemental Materials S1. These compounds are initially arranged in 96-well plates at a concentration of 10 mM, which are diluted to a concentration of 1 mM using dimethyl sulfoxide (DMSO) for testing against N. fowleri. DMSO was not exceeded at 1% of the total volume to avoid toxic effects on the parasites.
After a first screening, four pure compounds were purchased. Terconazole was obtained from Cymit Química S.L. (Barcelona, Spain). ABT-239 and PD-144418 were purchased from MedChemExpress (Monmouth Junction, NJ, USA). Clemastine was purchased from Sigma-Aldrich (Madrid, Spain).

Compound Library Preparation and In Vivo Testing

The FDA Pharmakon compound library was purchased from MicroSource Discovery Systems, Inc. (USA). Compounds (10 mM, dissolved in DMSO) were stored at -80 °C until use. Levamisole and ivermectin, used for the in vitro controls, were purchased from Fluka and Sigma-Aldrich, respectively (Buchs, Switzerland), dissolved at 10 mM stock solutions in DMSO, and stored at -20 °C until further use. For the in vivo studies, bitoscanate, chlorcyclizine HCl, clemastine, dicyclomine HCl, drofenine HCl, ethopropazine HCl, lansoprazole, metformin HCl, morantel, natamycin, trichlorfon, and trimipramine maleate were purchased from Sigma-Aldrich.
