Fitc conjugated anti p selectin

Manufactured by BD
Sourced in United States

FITC-conjugated anti-P-selectin is a fluorescently labeled antibody that binds to the P-selectin cell surface receptor. P-selectin is involved in cell adhesion and plays a role in inflammatory processes.

1 mM calcium was added to gel-filtered platelets. For flow cytometric assays, Alexa-Fluor-488–conjugated fibrinogen (Molecular Probes, Eugene, OR, USA) or FITC-conjugated anti–P-selectin (BD Pharmingen) were added with a range of concentrations of AYPGKF, a PAR-4 agonist peptide. Platelets were stimulated for 10 min at 37°C, and fixed for 10 minutes with 2% paraformaldehyde at RT. Washed platelets were diluted in Tyrode’s buffer and stained with FITC-anti-CD41, PE-anti-GPVI, and APC-anti-CD49b for 15 min before flow cytometric.

Thrombin, ADP and luciferase were bought from Chrono-Log Corporation (Havertown, PA, USA). FeCl3 was obtained from Sigma-Aldrich (St. Louis, MO, USA). FITC-conjugated anti-P-selectin and FITC-conjugated annexin V were purchased from BD Biosciences (Franklin Lakes, NJ, USA), while PE-conjugated Jon/A was purchased from Emfret Analytics (Würzburg, Germany). PRP was prepared in sodium citrate and stimulated with thrombin (in the presence of 10 µM Gly-Pro-Arg-Pro-NH2), CRP, ADP, or U46619 for 20 min at 37 °C in the presence of FITC-conjugated anti-P-selectin (BD Biosciences), PE-conjugated JON/A (Emfret, Würzburg, Germany) and APC/Cy7-conjugated anti-CD63 (Biolegend) antibodies. Platelets were subsequently fixed and analyzed by fluorescence-activated cell sorting (FACS) using an LSRFortessa cell analyzer (BD Biosciences) and FlowJo software.
For receptor expression studies, PRP was incubated with FITC-conjugated antibodies directed against surface membrane glycoproteins GP1b (CD42b), GPVI, integrin α2 (CD49b) (Emfret, Eibelstadt, Germany), integrin αIIb (CD41) (BD Biosciences, Oxford, UK), or PE-conjugated antibodies against integrin β2 (CD18) (Biolegend). Receptor expression was also studied upon stimulation with 0.1 U/mL thrombin for 20 min at 37 °C in the presence of 10 µM Gly-Pro-Arg-Pro-NH2. Platelets were subsequently analyzed by FACS.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
