Bacterial Binding Assay with CD33-Fc

Immulon 4HB ultrahigh binding microtiter plates (Thermo Scientific) were coated with 0.5 mg/mL protein A in coating buffer at 4°C overnight. Wells were washed three times and blocked with assay buffer. Human CD33 rSiglec-Fc chimeras (R & D Systems) were added to wells at 0.5 mg/mL and adhered at 4°C overnight. Wells were washed three times with assay buffer. 107 FITC-labeled bacteria suspended in 50μL assay buffer were added into the well and incubated at 37°C for 10 min. After washing, the fluorescent intensity (excitation 488 nm, emission 530 nm) was measured using a SpectraMax Paradigm Multi-Mode Detection Platform (Molecular Devices).
