Delipidation assays were performed by reacting 3 μg of synthetic peptides (synthesis described in supplementary information ) with 1μl of enzyme (hNotumcore or hNotumcoreS232A; 25 ng/μl) in 20 mM ammonium bicarbonate buffer (Total vol. 5 μl) for 16 hours at 25°C. The reaction was quenched with 0.1% TFA and samples were desalted using c18 zip tips. Samples were prepared in α-cyano-4-hydroxycinnamic acid in 50:50 water/acetonitrile with 0.1% TFA. MALDI-TOF spectra were acquired using an ABSCIEX 5800 TOF/TOF systems and analysed using data explorer v4.11.
