A total of 1 million passage 3 cells were resuspended in 200 µL of PBS containing fluorescein isothiocyanate– or phycoerythrin-coupled antibodies against CD34, CD44, CD45, CD90, CD147, CD271 (BD Biosciences), CD117 (eBioscience), CD105, CD166 (Ancell), and vascular endothelial growth factor receptor–2 (VEGFR-2; R&D Systems) and incubated for 30 minutes at 4°C. The cells were then washed with PBS and resuspended in 350 µL of PBS for analysis. Fluorescein isothiocyanate– or phycoerythrin-coupled nonspecific mouse Immunoglobulin G (BD Biosciences) was used as an isotype control. Cell fluorescence was evaluated via flow cytometry using an EC800 instrument (SONY), and data were also analyzed using the EC800 instrument.9 (link),23 (link),24 (link),32 (link)

Comprehensive Immunophenotypic Analysis of Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
One million cells at passage 2 were resuspended in 200 mL of phosphate-buffered saline solution (PBS) containing antibodies coupled with either fluorescein isothiocyanate or phycoerythrin. Coupled antibodies to cluster of differentiation (CD)31 (endothelial cell marker), CD34 (hematopoietic progenitor cell antigen), CD44 (hyaluronan receptor), CD45 (hematopoietic cell marker), CD90 (Thy1), CD147 (neuregulin; BD Bioscience, San Diego, CA), CD117 (C-kit, stem cell factor receptor; eBioscience, San Diego, CA), CD105 (SH-2) and CD166 (ALCAM, SB10; Ancell, Bayport, MN) were used. The vascular endothelial growth factor receptor 2 (VEGFR2) was obtained from R&D Systems (Minneapolis, MN). After incubation for 30 minutes at 4 C, the cells were washed with PBS and then resuspended in 350 mL of PBS for analysis. As isotype control for the primary antibody, fluorescein isothiocyanatee or phycoerythrin-coupled nonspecific mouse IgG (BS Biosciences) was used. Cell fluorescence measurements through flow cytometry and data analysis were carried out using an EC 800 instrument (SONY, Tokyo, Japan). 9, 10

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
