Sprague-Dawley Rat Inflammation Model

A total of 25 male Sprague-Dawley rats (4 weeks old, weighing 80-100 g, Samtaco Bio) were used in the present study. The animals were housed in a controlled environment with a temperature of 24±1°C and a humidity of 50±5% with a 12 h light and dark cycle. Food and water were provided ad libitum. The rats (n=5 in each group) were intraperitoneally injected with CA (50 mg/kg). After 3 h, LPS (0.5 mg/kg) was inoculated subcutaneously into the right flanks of the rats. After 24 h, the rats were sacrificed, subcutaneous tissues were excised, and then fixed in 10% buffered formalin and embedded in paraffin. Tissue sections (4-µm-thick) were stained with Hematoxylin (Sigma-Aldrich; Merck KgaA) for 5 min and eosine (Sigma-Aldrich; Merck KgaA) for 10 sec at RT and then examination by light microscopy.

Histological Analysis of Zebrafish Liver

Liver samples taken from 1.5, 3, 4.5, 6, and 7.5 days post-DOX treatment (dpt) fish were slowly dehydrated using a series of increasing ethanol solution concentrations (70%, 90%, 95%, and 100%). Specimens were then embedded in paraffin using a Leica EG1120. Sectioning at 5 μm was performed using a Reichert-Jung 2030 microtome. Hematoxylin (H) (Vector Laboratories, Burlingame, CA, #H3404) and Eosin (E; Sigma, #HT110232) were used for H and E staining. Oil-Red-O (Sigma) staining of zebrafish liver sections was performed as previously described [18 (link)]. Identification and classification of tumor types were based on previously established criteria [6 (link)].

Virgin Coconut Oil Supplementation in SHR Rats

SHR + coconut oil and SHR + trained + coconut oil groups were supplemented daily with virgin coconut oil in nature (2 mL•day -1 ). This dose was based on Nandakumaran et al. (2011) (link). Virgin coconut oil was administered by gavage and saline solution was used as vehicle. Dihydroethidium (DHE) was from Invitrogen (Carlsbad, Calif., USA). Hematoxiline and eosine were from Sigma-Aldrich (St. Louis, Mo., USA). Ketamine and xylazine were from Vetnil (Curitiba, PR, Brazil). Phenylephrine hydrochloride (PHE) and sodium nitroprusside (SNP) were from Sigma-Aldrich.
