Transgenic Col2-rtTA-Cre;Ext1e2fl/e2fl [Tg(Col2a1-rtTA,tetO-cre) 22Pjro; Ext1tm1.1Vcs]6 (link), R26R-LacZ [Gt(ROSA)26Sortm1Sor]17 (link); Ndst1fl/fl mice [Ndst1tm1Grob]15 (link), Prx1-Cre [Tg(Prrx1-cre)1Cjt]18 (link) and Col2-Cre [Tg(Col2a1-cre)1Star]19 (link) mice were maintained on a C57Bl/6J genetic background. Up to six females, one breeding pair or single males were kept in air filtered cages (SPF conditions, Green Line, Techniplast) under a 12 h light/dark cycle on regular bedding with nesting material (Abed) as environmental enrichment and food (Sniff) and water ad libidum.
Genotyping was performed by PCR on tail tip DNA (DirectPCR Lysis Reagent; VIAGEN) with primers listed in Table S1. Allele recombination in Col2-rtTA-Cre;Ext1e2fl/e2fl mice was induced by peritoneal injection of 80 mg Doxycycline per kg body weight (kgbw) into lactating dams at postnatal day 8 (P8)6 (link). Mice undergoing surgery received 1 mg/kgbw Doxycycline at 6 weeks.

Housing and Acclimation of C57BL/6N and BALB/c Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Male C57BL/6NCrl (B6N) and BALB/cAnNCrl (BALBc) were obtained from Charles River (Bad Sulzfeld, Germany) at an age of 6 weeks and housed in groups of 3–4 in IVC cages (Greenline, Techniplast) under SPF conditions (23°C ± 4°C and 50% humidity ± 10%) with food and water ad libitum and a 12 h:12 h light-dark cycle (lights on: 08:00 h). Experiments were performed during the light phase of the light-dark cycle (i.e., between 09:00 and 17:00 h). The cages were equipped with bedding and rodent tunnel (4.5 cm × 4 cm, diameter: 30 mm; ABEDD, Vienna, Austria). Animals had at least 10 days to recover from transportation and to adapt to the new holding conditions before the experiments started. Animals of a given experiment were delivered and tested at the same time in order to keep environmental factors constant. In case of 1H-MRS imaging and WCM training, mice were isolated at least 1 week before starting the experiments and individually housed thereafter. Otherwise, mice were kept in groups of 3–4. All experimental procedures have been approved by the Government of Upper Bavaria (ROB-55.2-2532.Vet_02-17-224). All efforts were taken to reduce animal numbers and to minimize animal suffering.

Animal Models for Infectious Disease Research

One-day-old chickens, 1-day-old turkeys, ferrets as well as C57BL/6 (B6) mice and human MxA transgenic (hMxAtg/tg) mice were used for this study. Chicks were bread at the FLI from SPF-chicken eggs (VALOBioMedia, Germany) and 1-day-old turkeys were ordered and shipped on hatching day from a local breeding facility (Bösel) to the FLI. The ferrets were obtained from the in-house breeding program at the FLI. B6 mice were obtained from Janvier and hMxAtg/tg mice were bred in-house at the Institute of Virology, Freiburg. All mice were housed in individually ventilated cages (Green Line, Tecniplast) with stable bedding (2HK: TAPVEI® BEDDING), at a temperature of 20.2 ± 1.1 °C, a humidity of 55% ± 7.1% and a 12 h:12 h light:dark cycle. The mice were fed ad libitum with food made from natural ingredients (Germany, altromin 1314). All animal work was performed in BSL3 containment facilities.

Murine Model for Networked Cage Housing

Check if the same lab product or an alternative is used in the 5 most similar protocols
Female C57Bl/6J mice (n = 60) were obtained from Charles River Laboratories (France). They were housed at a 12/12 light–dark cycle, and food and water were available ad libitum, first in conventional cages until the age of 11 weeks, or single-housed after implanting catheter and EMG electrodes to avoid implant damage by cage-mate biting. Cages with inserted dividers (green line, Tecniplast, Buguggiate, Italy) were used, allowing interaction by sound and smell and reducing social deprivation, complying with the 3R principles of animal experimentation.

Housing Conditions for NOD.Cg-Prkdc Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
NOD.Cg-PrkdcscidIl2rgtm1Wjl/SzJ mice were purchased from Charles River. All animal experiments were performed in accordance with protocols approved by the Service de la Consommation et des Affaires Vétérinaires of Canton de Vaud, Switzerland (VD 1865.3, 1865.4, and 1865.5). Mice were maintained and handled according to Swiss guidelines for animal safety with a 12-h-light-12-h-dark cycle, controlled temperature and food and water in polysulfone bottles ad libitum in SPF conditions, cages enriched with nesting material, cardboard, and wood tunnels, 12 h light cycle, 7 a.m. to 7 p.m. Animals are housed in IVC polysulfone cages—Green line—from Tecniplast®, type II long. Bedding is provided by Aspen Tapvei® (little squares about 4 mm × 4 mm × 1 mm). Water is acidified (pH between 2.5 to 3) on a resin column (Prominent® CH system). Diet from Provimi-Kliba® (cat# 3242, irradiated). Housing room temperature is 22 ± 2 °C, humidity 55 ± 10%.

Housing Conditions for Mouse Studies

Check if the same lab product or an alternative is used in the 5 most similar protocols
Animals used in this study were group-housed with two to five mice in individually ventilated cages (Tecniplast Germany GmbH, Green Line or Blue Line) and kept under identical housing conditions: 12h light/dark cycle (light cycle 6:30 a.m. to 6:30 p.m.), standard pelleted mouse diet (ssniff GmbH, Soest, Germany, article number v1124-300) ad libitum, free access to water, 22 ± 2°C room temperature and relative humidity of 55 ± 10%. The cages contained wooden bedding material (Tapvei Estonia OÜ, Estonia, Aspen bedding, article number 4HK 10 kg), nestlets (ssniff GmbH, Soest, Germany, article number H3279-10), a red plastic house (ZOONLAB GmbH, Castrop-Rauxel, Germany) and paper tunnels (ZOONLAB GmbH, Castrop-Rauxel, Germany; article number 3084030) as cage enrichment. Animals were handled by male and female caretakers and technicians.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
