Enzymatic Assay for Prostaglandin Synthesis

A total of 1 mg of purified proteins (TcAKR, LiPGFS1, LiPGFS2, TbPGFS, HsPGFS and TcOYE) were incubated in the enzyme assay buffer in the presence of 10 µM prostaglandin H2 (Cayman Chemicals) and 100 µM NADPH for 1 h at 37°C. Samples were inactivated by the addition of two volumes of acetonitrile and analysed by LC–MS/MS as described below for the analysis of arachidonic acid and a prostaglandin standard mixture (Cayman Chemicals).
