Hypothermia Effects on Blood Glucose and Norepinephrine

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 1 h reperfusion following tMCAO, mice were subjected to hypothermia for 4 h achieved by surface cooling, chemogenetic induction, or DBS stimulation. Then the blood was obtained by pricking the tail. The blood glucose level was measured using a glucometer (Roche, Accu-Check, USA). Additionally, the blood (100–200 μL) was obtained by penetrating the retro-orbital sinus in mice with a sterile haematocrit capillary tube and placed in an anticoagulant tube. After centrifugation at 3000 rpm for 5 min, the plasma was used for the detection of the noradrenaline level with an ELISA kit (CSB-E07870m, CUSABIO, China), following precisely the manufacturer’s recommendations.

Quantification of Serum and CSF Biomarkers

Check if the same lab product or an alternative is used in the 5 most similar protocols
The concentrations of transmitters and hormones in the collected samples of serum or cerebrospinal fluid were quantified using ELISA kits, specifically for cortisol (CUSABIO, E05113m, Wuhan, China, CSB-E05113m), NE (CUSABIO, CSB-E07870m), renin (CUSABIO, CSB-E08703m), and glutamate (Abnova, KA1670, Taiwan, China, KA1670). All experiments are performed according to the manufacturer’s instructions.

Serum Biomarkers Analysis Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
The venous blood samples were immediately collected and allowed to clot for 30 min at 24–25 °C before centrifuging at 2000× g for 15 min at 4 °C. Serum was transferred to a new tube and stored at −80 °C. We used ELISA kits for the analysis of the serum concentrations of leptin (KMC2281, Thermo Fisher Scientific, Waltham, MA, USA), noradrenaline (CSB-E07870m, CUSABIO, Wuhan, China), and insulin (80-INSMS-E01, ALPCO, Salem, NH, USA) and colorimetric kit for the detection of the serum concentration of glucose (K039-H1, Arbor Assays, Ann Arbor, MI, USA).

Profiling Serum Biomarkers in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected from 8-week-old mice by 1.5 mL centrifuge tubes at room temperature and allowed to stand still for 1h. Serum were collected from the upper layer after clotting and centrifuged at 3000 rpm for 10 min at room temperature. The protein levels of S100A8, S100A9, PINP, CTX-I, leptin and noradrenaline in the serum were measured using a mouse S100A8 ELISA kit (E-EL-M1343c, Elabscience, Wuhan, China), a mouse S100A9 ELISA kit (E-EL-M3049, Elabscience),a mouse PINP ELISA kit (E-EL-M0233c, Elabscience), a mouse CTX-I ELISA kit (CEA665Mu, cloud-clone, Wuhan, China), a mouse leptin ELISA kit (E-EL-M3008, Elabscience) and a mouse noradrenaline ELISA kit (CSB-E07870m, CUSABIO, Wuhan, China) according to the manufacturer’s instructions.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
