Immunofluorescence Staining of G3BP1

Cells were fixed with 4% paraformaldehyde in PBS for 15 min, permeabilized with 0.5% Triton X-100 in PBS for 10 min, and then blocked with 3% goat serum in PBST (PBS plus 0.1% Triton X-100) for 1 h, with all steps performed at RT. Cells were further incubated with anti–G3BP1 antibody (BD, 611127) overnight at 4 °C. AlexaFluor-488 (Invitrogen, A11001) used as the second antibody was incubated with the cells for 1 h at RT. After 3 washes with PBST, cells were mounted on glass slides using the ProLong Gold Antifade Mountant with DAPI (ThermoFisher, P36935). Images were captured using the Leica TCS SP8 confocal microscopy system with a ×100 objective (oil immersion). Images were processed and assembled into figures using LAS X (Leica) and Fiji.
