Peroxidase labeled secondary antibodies

Manufactured by Proteintech

Peroxidase-labeled secondary antibodies are conjugated with the enzyme horseradish peroxidase. They are used to detect and amplify the signal of primary antibodies in various immunoassays, such as Western blotting, ELISA, and immunohistochemistry.

Cells were harvested in phosphate-buffered saline (PBS) and centrifuged for 1 minute at 13,000 rpm. The pellet was resuspended in 100 μL of lysis buffer (radio-immunoprecipitation assay [Beyotime, Shanghai, People’s Republic of China], Cocktail, and phenylmethanesulfonyl fluoride [Hoffman-La Roche Ltd., Basel, Switzerland]) and incubated for 30 minutes on ice. The lysate was centrifuged for 10 minutes at 12,000 rpm at 4°C. The protein amount was determined using the bovine serum albumin (BSA) kit (Beyotime). For immunoblotting, the following antibodies were used: anti-MAP2K7 (55030-1-Ap; Proteintech, Rosemont, IL, USA), anti-E-cadherin (ab1416; Abcam, Cambridge, UK), anti-N-cadherin (4061; cell signaling technology), and anti-GAPDH (AP0063; Bioworld Technology, St Louis Park, MN, USA). Bound antibodies were detected by using peroxidase-labeled secondary antibodies (Proteintech) and immunoblotting and enhanced chemiluminescence detection reagents from Thermo Fisher Scientific.

Five micrograms of protein were electrophoresed in 10% SDS-PAGE gels and blotted to polyvinylidene difluoride membranes. Specific primary antibodies were detected with peroxidase-labeled secondary antibodies (ProteinTech Group Inc.) by using SuperSignal West Dura Extended Duration Substrate (Pierce Chemical) per the manufacturer’s instructions. The antibodies used from ProteinTech Group Inc. included the myc-tag antibody (Cat. #66036–1-Ig), ASS antibody (Cat. #66036–1-Ig), GAPDH antibody (Cat. #60004–1-Ig), FTL antibody (Cat. #10727–1-AP), Flag-tag antibody (Cat. # 66008–3-Ig), p53 antibody (Cat. #60283–2-Ig), Bcl-2 antibody (Cat. #60178–1-Ig), PUMA antibody (Cat. # 55120–1-AP), Bax antibody (Cat. #60267–1-Ig), caspase 9 antibody (Cat. # 66169–1-Ig), caspase 3 antibody (Cat. # 66470–2-Ig), Histone H3 antibody (Cat. # 17168–1-AP), HRP-conjugated goat anti-mouse IgG (Cat. #SA00001–1) and HRP-conjugated goat anti-rabbit IgG (Cat. #SA00001–2). The p53AIP1 antibody (Cat. # ABP56144) was supplied by Abbkine Inc., while the Noxa antibody (Cat. # ab13654) and the Bak antibody (Cat. # ab69404) were both from Abcam Inc. The TRITC conjugated goat anti-rabbit antibody (Cat. # AS10–1018) was from Agrisera Inc.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
