PHBV/Bacterial Cellulose Nanocomposite Fabrication

Poly-(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) with 12% poly-(3-hydroxyvalerate) content was supplied by Goodfellow Cambridge Ltd. (Huntingdon, UK) in granular form with a density of 1.250 g cm−3 and served as a matrix. The TES blowing agent was purchased from Nouryon (Sundsvall, Sweden) under the trade name of Expancel 920 DU 120. The plasticizer, acetyl tributyl citrate, was supplied by Sigma Aldrich (St. Louis, MO, USA).
Fresh bacterial cellulose pellicles containing about 1% cellulose and 99% water [31 (link)] were produced under static conditions by a Komagataeibacter xylinus strain and mechanically disintegrated as previously reported [26 (link)]. Briefly, bacterial cellulose nanofibers, denoted as BC, were obtained by defibrillating the pellicles with a high-speed blender for 30 min and, after being diluted with distilled water (1:1, w/w), with an LM20 Microfluidizer (Microfluidics, Westwood, MA, USA) for twelve passes. The resulting BC suspension in water containing about 0.5 wt% BC was frozen at −20 °C for 48 h and freeze-dried using a FreeZone 2.5 L equipment (Labconco, Kansas City, MO, USA).

Synthesis and Characterization of Fluorinated Surfactants

Tributyl O-acetylcitrate or acetyl tributyl citrate (ATBC), acetonitrile, and methanol were purchased from Sigma-Aldrich Chemical (Saint-Quentin Fallavier, France). Paclitaxel was obtained from Chemieliva Pharmaceutical Co., Ltd. Dulbecoo's minimal essential serum, fetal bovine serum and penicillin streptomycin, trypsin (1X) and trypan blue were purchased from ThermoFisher Scientific (Gibco, France). The CT26-murine colorectal carcinoma cell line was purchased from ATCC. Fluorinated surfactants, F n d i TAC m , possess a fluorinated chain length of n = 6 carbons with a double polar chain (i = 2) made of a repetition of m = 12 tris(hydroxymethyl)aminomethane (TAC) water soluble units. F 6 d 2 TAC 12 is a member of a recently developed new class of biocompatible branched surfactants called "Dendri-TAC" 26 . Their convergent synthesis allows stepwise modifications of the surfactant's size, shape, hydrophilic/lipophilic balance, nature of the head, number of tails, spacers or connecting units between hydrophilic head and hydrophobic tail. Their highly versatile structure affords not only to control the formation of structurally well-defined macromolecules but also to adjust step-bystep their self-assembling properties.
