Quantifying PC12 Cell Viability

PC12 cell viability was determined using a Cell Counting Kit-8 (CCK-8) cell viability assay (Dojindo Molecular Technologies, Inc., Kumamoto, Japan). PC12 cells were incubated in 96 well plates with 10,000 cells per well for cell viability analysis. Following the final 24 h incubation step of OGD/R or IPOC treatments, 20 µl CCK-8 solution was added to each well containing 200 µl culture medium. After incubating the cells at 37°C for 2 h, the absorbance at 450 nm (A450) was measured using a Multiskan FC microplate spectrophotometer (Thermo Fisher Scientific, Inc.). HUVEC Viability Assay with Lenvatinib

HUVECs (3×103 cells per well) were seeded in flat-bottomed 96-well plates. After incubation for 24 h, the cells were treated with lenvatinib at various concentrations and 60 ng/ml VEGFA165 (PeproTech Inc.) in serum-reduced (1%) RPMI 1640 medium for 72 h. The final concentration of dimethyl sulfoxide (DMSO, as vehicle) was ≤1%. Results were expressed as the mean ± S.E.M. of at least three independent proliferation assays. The dose-response curve and half maximal inhibitory concentration (IC50) values of lenvatinib were calculated busing the GraphPad Prism software (GraphPad Software Inc., San Diego, CA, USA).
