Fasting-Induced Hypoleptinemia in Mice

We have previously shown that 48 hr fasting and 2 week caloric restriction elicit similar undernourished phenotypes with weight loss, decreased leptin, and altered T cells (14 (link)). Therefore to model the state of undernutrition and induce hypoleptinemia, mice were fasted for 48 hours with access to water only (14 (link)). Following the 48 hr fasting period, mice were permitted to feed ad libitum. At the end of the fasting period, serum was assayed for leptin levels by ELISA using a Mouse/Rat Leptin Quantikine ELISA Kit (R&D Systems, Minneapolis, MN, USA).
Recipient baseline weights were recorded immediately prior to transplantation, prior to initiation of fasting, and again after the fasting period, which resulted in at least 25% weight loss (14 (link)). Fed mice were also weighed at these time points.
Injection of leptin in fasted mice was used as a “rescue” of the fasted state (14 (link)). Mice received IP injections of 25 µg of recombinant mouse leptin protein (R&D Systems) in 200 µl of 20 mM tris base, pH 8.0. Mice were injected twice daily during the fasting period, and mice not receiving leptin rescue received a sham rescue by IP injection of 200 µl of 20 mM tris buffer without leptin.
