Quantification of Isoniazid in Hair

We purchased reference standards for isoniazid (98% chemical purity) and isoniazid-d4 (internal standard, 98% chemical purity, 98.4% isotopic purity) from Toronto Research Chemicals (Ontario, Canada). We obtained analytical grade water, methanol, and acetonitrile from Honeywell Burdick and Jackson (Muskegon, MI). We prepared stock solutions of the standard and internal standard at 1 milligram (mg)/milliliter (mL), aliquoted to 1mL portions in amber vials and stored at -80°C. All calibration standards, ranging in concentration from 0.05 to 50 nanograms (ng)/mg hair, were prepared from the stock solution.

Analyte‐free human plasma (blank plasma) with potassium EDTA as an anticoagulant was obtained from healthy and untreated volunteers (men and women aged 25–30 years).

Quantitative Determination of Isoniazid and Acetyl-Isoniazid

Reference standards for isoniazid (97.1 % purity), acetyl-isoniazid (98.0 % purity), isoniazid-d4 (INH-d4; used as internal standard 98.0 % purity), and acetyl-isoniazid-d4 (AcINH-d4; used as internal standard 98.0 % purity) were purchased from Toronto Research Chemicals (TRC).
Ammonium acetate and dimethyl sulfoxide (DMSO) (high purity grade) were purchased from Sigma-Aldrich (Darmstadt, Germany). Ammonium bicarbonate (high purity grade) was purchased from Acros organics (Geel, Belgium). Acetonitrile and methanol (LC-MS grade) were purchased from Honeywell (Burdick and Jackson, Muskegon, Michigan), and LC-MS grade water was produced in-house using the Milli-Q water purification system (Merck Millipore, Darmstadt, Germany). Strata X 33 µm Polymeric Reversed Phase 200 mg/3mL solid phase extraction (SPE) columns were purchased from Phenomenex (California, United States).

Quantitation of Anti-tuberculosis Drugs

Pyrazinamide (lot: 100178–201104), isoniazid (lot: 100578–201502), rifampicin (lot: 130496–201403), and ethambutol dihydrochloride (lot: 100165–201705) were purchased from National Institutes for Food and Drug Control. Internal standards (ISs), pyrazinamide-D3 (lot: CRC-OW3-056), isoniazid-D4 (lot: 2-JLO-114-1), rifampicin-D3 (lot: 5-YEN-103-103-1), rifampicin-D8 (lot: NJ-07222019), and ethambutol-D4 (lot: 1-WEW-51-4), were purchased from Toronto Research Chemicals, Canada. The purities of these analytes were all above 98.0%. Molecule structures of the four analytes and corresponding ISs are presented in Figure 1.
Methanol (HPLC grade) and acetonitrile (HPLC grade) were purchased from Merck Drugs & Biotechnology, German. Trifluoroacetic acid (HPLC grade) was purchased from Sigma Aldrich Fluka, USA. Ammonium formate (HPLC grade) was purchased from Anaqua Chemical Supply, USA. Water was prepared by a Milli-Q deionization system (Millipore, Billerica, MA, USA). Blank human serum was obtained from the physical examination population of The First Affiliated Hospital, Zhejiang University School of Medicine.

Quantification of Isoniazid Compounds

INH, AcINH, and isoniazid-d4 (INH-d4; used as an internal
standard) were purchased from Toronto Research Chemicals (North York,
Canada). Optima-grade LC–MS solvents of methanol, acetonitrile
(ACN), and water were used to prepare solutions and solvent mixtures
(Fisher Scientific, Leicestershire, UK). Formic acid was added to
the mobile phase solvents at 0.1% (v/v) (Fischer Scientific, Leicestershire,
UK). Dichloromethane was used to prepare solvent extraction mixtures
and was of analytical or reagent grade (Sigma-Aldrich, Dorset, UK).
A solution of artificial eccrine perspiration was used to simulate
a fingerprint matrix in the experimental design of fingerprint experiments
(Pickering Laboratories, Inc., Obertaufkirchen, Germany).

Acetonitrile and methanol were purchased from Honeywell (Charlotte, United States). LC-MS/MS grade millipore water was produced in-house (Merck-Millipore, Germany).
