UAS-dSLC5A5-FLAG transgenic flies were generated by BestGene Inc. (Chino Hills, CA, USA). For each cross, the Gal4 or UAS-RNAi progenies derived from the cross are used as controls.

Drosophila Genetics: Characterizing ski3 Mutants

Drosophila stocks were raised at 25 °C under a 12-h light and 12-h dark cycle, using a standard cornmeal-sugar-agar medium containing propionic acid and n-butyl p-hydroxybenzoate as mold inhibitors. To characterize ski3 mutants, w 1118 ;PBac {w +mC =WH}CG8777 f03251 /CyO, act-GFP (BDSC #18632; Bloomington, IN, USA) was backcrossed to our laboratory strain w 1118 (used as a control) for at least six generations prior to use in the experiments. arm-gal4 (BDSC #1561) and UAS-ski3-IR (NIG-FLY #8777R-1; Mishima, Japan) were used to knock down ski3. Mosaic clones were generated with an FLP/ FRT-mediated recombination system using the following stocks: DGRC #108449: w 1118 ;P{w +mW.hs =FRT(w hs )}G13P {w +mC =Ubi-GFP.nls}2R1P{w +mC =Ubi-GFP.nls}2R2, DGRC #141404: w*;P{w +mW.hs =FRT(w hs )}G13, y*w*;P{ry +t7.2 = neoFRT}40A P{w +mW.hs =FRT(w hs )}G13 cn 1 PBac{SAstop DsRed}LL04977 bw 1 /CyO, S* bw 1 , BDSC #6: P{ry +t7.2 = hsFLP}1, w 1118 ;Adv 1 /CyO. Heat shock was applied for 1 h at 37 °C to embryos at 0-6 h of age [13] . act-Gal4 (BDSC #3954) was used as a driver of UAS transgenes.
