Apoptosis Analysis of HK-2 Cells

HK‐2 cells were seeded in 6‐well plates (1.5 × 106 cells/well) and incubated for 24 hours. Next, the medium was replaced with DMEM/F12 with or without COM (300 µg/mL) and incubate for 48 hours. All cells in each well were collected and doubly stained with Annexin V‐FITC/propidium iodide (PI) or Annexin V‐APC/7‐ADD Apoptosis Detection kit (BD Biosciences) according to the manufacturer's instructions. Finally, the proportions of cell apoptosis were determined by flow cytometry (BD FACSCalibur). Three independent experiments were conducted in triplicate.

Annexin V-APC/7-AAD Apoptosis Assay

Apoptotic cells were detected using an Annexin V-APC/7-ADD apoptosis detection kit (BD Biosciences, USA). Briefly, cells were detached using EDTA-free trypsin, centrifuged, and the supernatant discarded. They were then rinsed with ice-cold PBS, centrifuged, and the cell pellet resuspended in 200 μl of 1 × binding buffer. The cells were then transferred into a FACS tube and stained with Annexin V-APC and 7-AAD (5µL each) in the dark for 15 mins, at room temperature. Next, flow cytometry measurements were taken, and the data analyzed on FlowJo 8.1.
