Isolation of Mouse Splenic DCs

DCs were isolated from the mouse spleen of naïve, ITD-treated, or heart transplant recipients. Briefly, spleens were digested with collagenase D (1 mg/mL; Boehringer Mannheim, Roche, Germany), and mononuclear cells were isolated by Histopaque 1083 (Sigma-Aldrich Inc., Saint Louis, MI) gradient centrifugation. Then, CD11c + cells were isolated using AUTOMACS and a mouse pan Dendritic Cell Isolation Kit (Miltenyi Biotec Inc.). Purity was >95% as determined by microscopy. Splenic CD8α + DCs and CD8α -DCs were also isolated by AUTOMACS and a mouse CD8α + Dendritic Cell Isolation Kit (Miltenyi Biotec Inc.).
