Hydrochloroauric acid trihydrate (HAuCl4∙3H2O), caffeic acid, 4-nitrophenol, 4-aminophenol, ninhydrin, acetic acid and NaBH4 were purchased from Sigma-Aldrich (St. Louis, MO, USA). Extraction of Crude Protein from Germinated Chestnuts

The crude protein solution was prepared according to the schematic diagram shown in Fig. S1. The germinated chestnuts (5.0 kg) were ground using a blender in 7.0 L of 100 mM Tris-HCl (pH 7.0). Four liters of n-hexane/diethyl ether (1:1, v/v) were added to the mixture, which was stirred at 4 °C for 1 h. Then, the mixture was centrifuged at 3000×g for 15 min at 4 °C to separate the lipid and aqueous layers. A separatory funnel was used to collect the aqueous phase, and the solution was centrifuged at 10,000×g for 30 min at 4 °C. The precipitate was then removed, and the supernatant was filtered through 0.45-μm Minisart RC syringe filters (Sartorius, Goettingen, Germany). The crude protein sample was collected into a bottle and stored at 4 °C for subsequent experiments.
