The Mouse VEGF Quantikine ELISA Kit is a quantitative sandwich enzyme immunoassay designed to measure mouse Vascular Endothelial Growth Factor (VEGF) levels in cell culture supernates, serum, and plasma. The kit uses a monoclonal antibody specific for mouse VEGF coated on a microplate to capture the VEGF present in the sample. An enzyme-linked polyclonal antibody specific for mouse VEGF is then added, and after a series of washes, a substrate solution is added to the wells and color develops in proportion to the amount of mouse VEGF bound.

Cytokine profiling of murine liver

Snap-frozen liver tissues were homogenized using the mixer mill MM 301 (Retsch, Haan, Germany) and subsequently reconstituted in PBS + 1% IGEPAL ® CA-630 (Sigma Aldrich), as well as a protease inhibitor cocktail (Sigma-Aldrich) in order to avoid proteolytic degradation. VEGF Secretion in Hypoxic PBMCs

Quantitative ELISA for Plasma and Tissue Proteins

The following commercially available ELISA kits were used to measure protein levels in plasma and tissue lysates: mouse VEGF Quantikine ELISA kit (R&D Systems; MV00), human sFlt Quantikine ELISA kit (R&D Systems; DVR100B), and mouse Epo Quantikine ELISA kit (R&D Systems; MEP00). Blood for plasma collection was drawn via the tail vein with 12 mM EDTA or heparin as an anticoagulant and then centrifuged for 20 min at 2,000 g. Tissue lysates for ELISA were homogenized in PBS using a Next Advance Bullet Blender homogenizer, stored overnight at −20°C, and lysed with two freeze–thaw cycles followed by centrifugation for 5 min at 5,000 g and collection of the supernatant. The Bradford assay was performed to standardize tissue lysate concentrations according to the manufacturer’s instructions (Bio-Rad Protein Assay Dye Reagent Concentrate; 500-0006). ELISAs were read at 450 nm with a reference value of 540 nm using a Tecan Infinite f200 Pro 96-well plate reader.

VEGF-A Secretion in Bbs4 Knockout Islets

Serum VEGF and TNF Quantification
