Lipid-based Nanoparticle Synthesis and Characterization

Lipids (1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC) and 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP)) were purchased from Avanti Polar (Alabaster, AL, USA). LAH4-L1 peptide (KKALLAHALHLLAL-LALHLAHALKKA) was purchased from GenScript (Piscataway, NJ, USA). Absolute anhydrous ethanol was purchased from Carlo Erba Reagents (Peypin, France), sterile pyrogen-free bidistilled water OTEC® was purchased from Aguettant (Lyon, France), and nuclease-free water was purchased from Ambion (Thermo Fisher Scientific, Waltham, MA USA). Furthermore, 1× and 10× DPBS (Dulbecco’s phosphate-buffered saline, pH 7.4), culture medium (RPMI/glutamax and DMEM), fetal bovine serum (FBS), N-2-hydroxyethylpiperazine-N-2-ethane sulfonic acid (HEPES), and β-mercaptoethanol were all purchased from Gibco (Dublin, Ireland). Lipofectamine 2000TM transfection reagent was purchased from Invitrogen™ via Thermo Scientific™ (Waltham, MA, USA).

Synthesis and Purification of PCL-b-PEG

The following analytical-grade chemicals were purchased and used without further purification: 2,3,5-triiodobenzoic acid (Changzhou Ilhang Fine Chemical Co. Ltd, China), thionyl chloride (Acros Organics, 99.5%), sodium carbonate, dichloromethane (Acros Organics, 99.9%). N-methyl-2pyrrolidone (NMP, 99.5%), pyridine (99.5%) and 4dimethylaminopyridine (DMAP, 99.5%) were purchased from Acros Organics in anhydrous grade over molecular sieves in an AcroSeal packaging. Poly(vinyl alcohol) (PVAL, Sigma-Aldrich, 98% hydrolyzed, average molar mass MW = 13,000-23,000 Da) was dried at 40 °C in an oven for 24 h under vacuum before use. Tetrahydrofuran (THF, Fisher Scientific), absolute anhydrous ethanol (Carlo Erba) were used in purification processes. The polycaprolactone-block-poly(ethylene glycol) (PCL-b-PEG block copolymer) was synthesized by polymerization of caprolactone initiated by poly(ethylene glycol) monomethyl ether catalyzed by tin octanoate as reported in refs 58, 59 The degrees of polymerization of the poly(caprolactone) and. poly(ethylene glycol) blocks were 65 and 113 respectively (PCL65-b-PEG113). Deionized water of 18 Ω•cm resistivity was used for all preparations.

Chitosan Characterization and FITC-Dextran Probes

Solvent Extraction and Chromatographic Analysis

Stock standard solutions were prepared in methanol at a concentration of 100 µg/mL and stored at −20 °C; working solutions were weekly prepared after a proper dilution.

Lipid Bilayer Preparation and Analysis

Milli-Q
water (18.2 MΩ cm) was used to prepare all solutions. Cardiolipin
(CL) solution from bovine heart (∼4.7–5.3 mg mL–1 in ethanol, ≥97%) and cytochrome c from equine heart (≥95%) were obtained from Sigma-Aldrich
and stored in a freezer (−18 °C). 1,2-Dioleoyl-sn-glycero-3-phosphocholine (DOPC) powder was purchased
from Avanti Polar Lipids and stored in a freezer. The 10 mM phosphate
buffer aqueous solution at pH 7.2 was prepared by using sodium hydrogen
phosphate anhydrous ACS (99.0% min) and potassium dihydrogen phosphate
ACS (99.0% min) from Alfa Aesar. Anhydrous absolute ethanol from Carlo
Erba Reagents was used to prepare DOPC/CL lipid mixture solutions.
Concentrated hydrochloric acid (12 M, 37%, ACS Reagent) and pellets
for making sodium hydroxide solution (ACS) were obtained from VWR
and were used to adjust the pH of the phosphate buffer solution. Sodium
nitrite (>99%) was obtained from Sigma-Aldrich, and 4-nitrocatechol
(>98%) was obtained from Alfa Aesar.

Quantitative Carbohydrate Analysis
