Oxidized LDL-Induced Macrophage Apoptosis

Wild type and lipin-1mKO bone marrow-derived macrophages were collected and resuspended in D1 media (DMEM; (Gibco 10829) 1% fetal bovine serum (Atlanta biologicals S11150), 2 mM glutamax (Thermofisher 35050–061), 100U/ml penicillin-streptomycin (ATCC), 1 mM sodium pyruvate (HyClone). Cells were placed in flow tubes (VWR 60818–500) and treated with 25 or 50 μg/ml of oxidized LDL for 24 h. LDL was oxidized as described previously [17 (link)]. After 24hrs, cells were centrifuged, washed with FACs wash, and stained with APC Annexin V Apoptosis Detection Kit with PI following the kit protocol (Biolegend 640932) and analyzed via ACEA NovoCyte Quanteon Flow Cytometer.
