Prophylactic and Therapeutic Interventions for Allergic Airway Inflammation

For prophylactic AA experiments, 2 mg of AA (Fisher Scientific: #ICN19462510) was diluted in 30 μL of corn oil (Sigma C8267) which was administered 4–6 hours prior to CS challenge via oral gavage. Control animals received 30 μL corn oil alone. All other experiments used saline controls. For exogenous Ang1 and Ang2 experiments, 500 ng of Ang1 or Ang2 (R&D Systems: #9936-AN and #7186-AN respectively) was administered via intraperitoneal injection 1 hour prior to CS challenge. For anti-Angiopoietin-2 antibody treatment 50 μg of antibody (Adipogen: #AG-27B-0016PF) was administered concurrently with CS challenge via intraperitoneal injection. For L-NAME experiments, 10 μg of L-NAME (Abcam: #120136) reconstituted in distilled water was injected via intraperitoneal injection concurrently with CS challenge. Finally, for L-Arginine experiments, 5 mg of L-Arginine (Cayman Chemical: #23703) reconstituted in distilled water was administered via intraperitoneal injection 4-6 hours prior to CS challenge. The schedules described above were maintained for all combinational interventions described.

Preparation of Compound Stock Solutions

100 mM stock solutions of all screened compounds were prepared in individual, sterile amber vials. All stock solutions were dissolved in sterile DMSO, except L-arginine, which was solubilized using dH2O, due to solubility constraints. Sterile cell-culture grade DMSO, tamsulosin hydrochloride, nifedipine, isoproterenol, butoxamine, adenosine and L-arginine were obtained from Sigma Aldrich. PGE1, PGE2, rolipram, L-arginine, ondansetron, celecoxib, diclofenac, mirabegron, sildenafil, atropine, vardenafil and tiotropium were obtained from Cayman Chemical. Y-27632 dihydrochloride was obtained from R&D Systems. Stock solutions when unused were stored at −20 °C

Glucose-6-Phosphate Quantification in Cells

HOS and U2OS cells were incubated with L-arginine (cat. no. A0013; Beijing Solarbio Science & Technology Co., Ltd.) and glucose-free F-12K (cat. no. 21127022, Gibco; Thermo Fisher Scientific, Inc.) medium for 30 min at 37°C and then incubated with 7 mM L-glucose (cat. no. 20829; Cayman Chemical Company), 1 mM L-arginine, or 1 mM D-arginine (cat. no. A769515; Toronto Research Chemicals) for another 30 min at 37°C. Cell lysates were then collected and glucose 6-phosphate content was determined using a Glucose-6-Phosphate Fluorometric Assay kit (cat. no. 700750-96 wells; Cayman Chemical Company). Finally, the concentration of glucose 6-phosphate in cells was determined according to the methods reported in the literature (27 (link)).
