NF-κB Reporter Assay with 4E-BP2

The NF-κB reporter (luc)-HEK293 cell line (BPS Bioscience) contains a firefly luciferase gene driven by four copies of NF-κB response element located upstream of the minimal TATA promoter. Stable cells were transfected with 4E-BP2 isoforms as above and supplemented with 100 ng pTK-RL (Gkogkas et al., 2008 (link)) for 48h and lysed 24h later (+/− TNFα 1nm) in lysis buffer from the Dual Luciferase Reporter Assay kit (Promega). Firefly and renilla luminescence were measured using a FLUOstar OPTIMA micro-plate reader (BMG LABTECH). Firefly luciferase luminescence values were normalized to renilla firefly luminescence values and are averages of four experiments.
