n-Hexane, ethyl acetate, and methanol (all HPLC grade) were obtained from Fisher Chemical (Argenta, Poznań, Poland). The peptides Bombesin and Orexin-A were from Cayman Chemical, and stock solutions were prepared in water and stored at −80 °C. Melatonin was purchased from Cayman Chemical or Tocris Bioscience, and stock solutions were prepared in dimethyl sulfoxide (DMSO); experimental samples contained a final concentration of 0.1% DMSO or less.

Assaying ESCC cell lines with compounds

Human ESCC cell lines (EC109, EC9706, KYSE150 and TE-1) and HEK-293T were obtained from the Hunan Fenghui Biotechnology Co., Ltd. (Changsha, China). These cells were cultured in high glucose DMEM medium (C11995500, Gibco, NY, USA) supplemented with 10% fetal bovine serum (04-001-1A, FBS, Biological Industries, Israel) and penicillin–streptomycin solution (100 U/ml, P1400, Solarbio, Beijing, China). Melatonin (#14427, Cayman Chemical Company, Michigan, USA), SAHA (HY-10221, MedChemExpress, USA), Lf3 (HY-101486, MedChemExpress, USA) and 10058-F4 (HY-12702, MedChemExpress, USA) stock solution was prepared in DMSO and diluted in culture media immediately prior to the subsequent experiments. The ESCC cells were treated with these chemical compounds for 48 h and then harvested for further analyses.

Rat Model of Cisplatin-Induced Neuropathy

In order to achieve a rat model of cisplatin-induced neuropathy, cisplatin (Item no: 13119, Cayman Chemical, Michigan, USA) was dissolved in saline and was intraperitoneally (i.p.) administrated at the dose of 3 mg/kg once a week for five weeks (total cumulative dose is 15 mg/kg), as previously reported in the literature24 (link),28 (link),29 (link). In order to prevent kidney injury due to cisplatin injection, the calculated dose was mixed with 2 ml of physiological saline prior to injection. Melatonin (Item no: 14427, Cayman Chemical, Michigan, USA) was dissolved in 5% ethanol and was administered to the rats in CP + MEL group intraperitoneally at a dose of 10 mg/kg/day for 15 consecutive days.

Puried deionized water with 18 MU cm conductivity was obtained using a Milli-Q water purication system (Millipore-Bedford, MA, USA). Whenever feasible, the analyte standards as shown in the ESI Table S1 † were purchased from two different sources in order to allow independent preparation of calibration and quality control samples. The vendor for stable isotope labeled internal standards as shown in the ESI Table S1 ] hydrochloride from Cayman Chemicals. Lastly, methanol (LC/MS grade) and acetonitrile (LC/MS grade) were purchased from Fisher Scien-tic (Göteborg, Sweden), along with formic acid (LC/MS grade), ammonia solution 25%, and dimethyl sulfoxide (DMSO) from Merck Life Science AB (Solna, Sweden).
