Preparation of Compound Stock Solutions

100 mM stock solutions of all screened compounds were prepared in individual, sterile amber vials. All stock solutions were dissolved in sterile DMSO, except L-arginine, which was solubilized using dH2O, due to solubility constraints. Sterile cell-culture grade DMSO, tamsulosin hydrochloride, nifedipine, isoproterenol, butoxamine, adenosine and L-arginine were obtained from Sigma Aldrich. PGE1, PGE2, rolipram, L-arginine, ondansetron, celecoxib, diclofenac, mirabegron, sildenafil, atropine, vardenafil and tiotropium were obtained from Cayman Chemical. Y-27632 dihydrochloride was obtained from R&D Systems. Stock solutions when unused were stored at −20 °C

Adrenergic Receptor Agonists: In Vivo, In Vitro, and Ex Vivo Applications

The pan β-adrenergic agonist, isoproterenol hydrochloride
(ISO) was from Sigma; the β3-adrenoreceptor agonists, CL
316,243 disodium salt (CL) was obtained from Tocris Bioscience (UK)
and mirabegron (Mira) was from Cayman Chemical. For CL
administration in vivo, mice were acclimated to
thermoneutrality for 3 days, injected with 1 mg/kg CL, and euthanized for tissue
collection 60 minutes later. For β-adrenergic stimulation in
vitro
, 3T3-L1 cells were treated with ISO (10 μM) for 40
minutes at 370C. For β-adrenergic stimulation ex
vivo
, human fat samples (approx. 0.5cm in size) were treated with
ISO (10 μM) for 40 minutes at 370C. For β-adrenergic
stimulation in vivo, human subjects were administered Mira for
10 weeks (50 mg/day), with abdominal adipose biopsies before and after
treatment, as described previously (Finlin
et al., 2018
).

Pharmacological Modulation of β3-Adrenoceptor

CL316,243 was purchased from Tocris and dissolved in water. CL316,243 was administered at one dose (1 mg/Kg/day) for seven consecutive days by interperitoneal injections (IP). Mirabegron was purchased from Cayman Chemical and dissolved in water and was administered at one dose (1 mg/Kg/day) for seven consecutive days by IP. Talinolol was purchased from Cayman Chemical and was dissolved in DMSO. Administration solution was dissolved further in water (DMSO ~5%) and was administered at one dose (1 mg/Kg/day) for five consecutive days by IP. SR59230A was purchased from Sigma-Aldrich and was dissolved in DMSO. Administration solution was dissolved further in water (DMSO ~5%) and was administered at one dose (1 mg/Kg/day) for five consecutive days by IP.

Insulin and β-Adrenergic Modulation

All cell culture consumables were from Thermo Fisher Scientific (Scoresby, Victoria, Australia). Human insulin (Actrapid) was obtained from Novo Nordisk. Mirabegron was obtained from Cayman Chemicals. CGP20712A, fatty‐free BSA, forskolin, ICI118551, (–)‐isoprenaline, and SR59230A were obtained from Sigma‐Aldrich. Rosiglitazone was obtained from Tocris Bioscience. All other reagents were of analytical quality.
