Quantification of Soluble c-Met in Plasma

The concentration of soluble Met in plasma was quantitated using human soluble c-Met quantitative ELISA kit (Invitrogen) which was described in previous study [9 (link)]. Briefly, 100 μL of plasma diluent was added into the well precoated with capture antibody and incubated at room temperature for 2 h. Then the medium was removed and the wells were washed with PBST buffer. 100 μL of detection antibody was added and incubated at room temperature for 2 h. The detection antibody was removed and wells were washed with PBST buffer. 100 μL of substrate reagent (R&D, DY999) was added and incubated at room temperature for 20 min. Then 50 μL of 5 M H2SO4 was added to stop reaction; the density of solution in wells was read at OD450 nm/570 nm, respectively. The concentration of s-Met was calculated using standard curve following the suggestion of manufacturer.
