Comprehensive qRT-PCR Analysis of Adipose Tissue

Total RNA was extracted from ATs with the Qiazol RNeasy Lipid Tissue mini kit (QIAGEN, Valencia, CA, USA) or with RNA-Stat 60 Reagent (Ams Biotechnology, Abingdon, UK) for 3T3-L1 cells, according to the manufacturer’s recommendations. Then, 1, 2, or 4 μg of total RNA was converted to cDNA using RT (Transcriptor Reverse Transcriptase 20 U/μL, 03531287001; Roche). Next, qRT-PCR amplification was performed using specific TaqMan Gene Expression Assays, 10 ng of cDNA, and Brilliant III Ultra-Fast qPCR Master Mix. Specific signals were normalized to the constitutively expressed TATA sequence binding protein (TBP; Mm 00446973; Thermo Fisher Scientific) for AT using the formula 2−Δct or to β-actin (mouse ACTB, 452341E; Thermo Fisher Scientific) for 3T3-L1 cells using the formula 2−ΔΔct. Trout Brain RNA Extraction and qPCR

Trout were anesthetized using 2-phenoxyethanol (1ml/L H2O) and then sacrificed by rapid decapitation. Brain tissue was collected, weighed, and immediately homogenized in QIAzol® Reagent (QIAGEN) for total RNA isolation using the QIAzol® RNeasy Lipid Tissue Mini Kit (QIAGEN) along with the RNase-Free DNase Set (QIAGEN) to remove genomic DNA. Whole brain tissue was collected for use in standard curve assays for rtPCR, as well as for the isolation of the trout rem2 gene fragment to serve as a cRNA probe template for in situ hybridization. For the relative expression levels of the rem2 gene in the major subdivisions of the trout central nervous system, brains were dissected from six individual animals and the olfactory bulb, cerebrum, diencephalon and midbrain, cerebellum, and hindbrain of each were isolated, as was a portion of the anterior region of the spinal cord.
