Cell Proliferation Assay with U0126

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were plated on laminin-coated 96-well plates at a density of 1000–3000 cells/well. At designated time points, CCK8 reagent (Dojindo, CK04-20) was added into each well according to manufacturer’s instructions. After incubation for 3–5 hours, OD450 absorbance of each well was screened by a microplate reader (Tecan, infinite M200PRO). For cell response to U0126, cells were plated out for 24 hours before U0126 was added into the wells.

Cell Growth Assay Using CCK-8

Check if the same lab product or an alternative is used in the 5 most similar protocols
Stably or transiently transfected cells were used for cell growth assays. The proliferation assay was performed in accordance with the manufacturer’s instructions (CCK8-Kit; CK04-20, Dojindo, Rockville, MD).

siRNA-mediated proliferation assay in H23 cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Dharmacon siRNA reagent (#G-005805-01) was delivered into H23 cell through Reverse transfection method67 (link). Briefly, siRNA and lipid (Lipofectamine® 2000 Reagent, 11668019) were mixed and added to 96-well plates. Then, H23 cells were added to the plates (10,000 cells per well). Cell proliferation rate was measured on day 3 by CCK8 assay (CK04-20, Dojindo Molecular Technologies, Inc.). During screening, siRNAs targeting EGFP (5′- CGTGATCTTCACCGACAAGAT-3′) are included in the upper-most and lower-most rows of each plate, which served as negative control. The screening was conducted once.

Anti-CD3 Antibody-Mediated T Cell Activation

Check if the same lab product or an alternative is used in the 5 most similar protocols
LEAF purified anti-human CD3 antibody (clone UCHT1, BioLegend, 300438) was immobilized (5 μg/ml) on an assay plate overnight at 4°C. After removal of unbound anti-human CD3 antibody, uncharacterized PBMC (C.T.L., CTL-UP1, lot no. HHU20180509) effector cells (3 × 104 cells per well), indicated doses of purified antibodies, and HCC1954 target cells (1 × 104 cells per well) were added and incubated for 72 hours at 37°C in a humidified incubator with 5% CO2. Supernatants were collected and assayed for IFN-γ secretion using a human IFN-γ Quantikine ELISA kit (R&D Systems, SIF50). Cell viability was analyzed by measuring absorbance at 450 nm after adding cell counting kit-8 (Dojindo, CK04-20) according to the manufacturer’s protocol.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
