Erythroid Cell Development Analysis

Comprehensive Stem Cell Marker and Integrin Profiling

Cells were first stained with fixable viability dye e780 (e-bioscience) according to manufacturer's instructions. Cells were then fixed with 4% paraformaldehyde (PFA) for 15 min prior to incubating the cells with the following conjugated antibodies to test for the expression of known stem cell markers: CD90 PE-Cy7 (e-bioscience), CD105 PE (ebioscience), CD44 APC (e-bioscience), CD73 e450 (e-bioscience), TRA-1-60 PerCP e710 (e-bioscience), OCT4 e660 (e-bioscience).
Additionally, the following antibodies were used to characterize the expression of integrin binding proteins: CD49b BV421 (BD Bioscience), CD29 BV510 (BD Bioscience), CD61 FITC (e-bioscience), CD49e PE (ebioscience), CD104 e660 (e-bioscience), CD49a PerCP e710 (e-bioscience) and CD49f PE-Cy7 (e-bioscience). All samples were run and analyzed on a BD LSRII flow cytometer using BD FACSAria.
