About 1 μg of ABCA3 protein was denatured and treated with 0.3 μl of PNGase F (P0708S, New England Biolabs) in 20 μl of reaction mixture at 37°C for 1 hour before being applied to SDS-PAGE and transferred to polyvinylidene difluoride membranes (Millipore). After blocking with 5% milk [in PBST, phosphate-buffered saline (PBS) with 0.1% Tween 20] at room temperature (RT) for 1 hour, the membranes were incubated with primary mouse anti-Flag (D191041, Sangon Biotech) or mouse anti-His (D191001, Sangon Biotech) monoclonal antibodies (1:5000 dilution in PBST) at RT for 2 hours. Following three rounds of 5-min wash in PBST, the membranes were incubated with horseradish peroxidase–conjugated goat anti-mouse immunoglobulin G antibodies (1:7000 dilution in PBST; D110087, Sangon Biotech) at RT for 1 hour and developed with UltraSignal hypersensitive ECL chemiluminescence substrate (4AW011, 4A Biotech) on an Amersham Imager 600 (GE Healthcare).
