Synthesis of PDMS-based Hydrogels

Sylgard 184 polydimethylsiloxane
(PDMS) was purchased from Dow Corning Corporation. Poly(ethylene glycol)
diacrylate (PEGDA, Mn ∼ 575 g/mol),
poly(ethylene glycol) dithiol (PEGDT, Mn ∼ 1000 g/mol), 2,2-dimethoxy-2-phenylacetophenone (DMPA or
Irgacure 651), 2-hydroxy-4′-(2-hydroxyethoxy)-2-methylpropiophenone
(Irgacure 2959, 98%), and dichloromethane (99.5%) were obtained from
Sigma-Aldrich. Poly(mercaptopropylmethylsiloxane) (PDMS-SH) (Mn = 4000–7000 g/mol; mercaptopropyl siloxane
unit content is 100%) and α,ω-vinyl-terminated PDMSs (vinyl-PDMS; Mw 28,000, 1000 cSt) were obtained from ABCR
GmbH, Germany. Sodium carboxymethylcellulose (CMC, DP 1100, Mw 250,000, and degree of substitution, 0.7)
was provided by GlaxoSmithKline UK. Phosphate buffer saline (PBS)
solution was prepared by adding a tablet of PBS in 200 mL of deionized
water. Photoirradiation was carried out using an Omnicure series 1500
lamp, and an ILT 1400-A radiometer photometer from International Light
Technologies was used to measure the corresponding irradiation intensities.

Synthesis and Functionalization of Gold Nanoparticles

Gold chloride trihydrate (HAuCl4∙3H2O), sodium borohydride (NaBH4), chlorhydric acid (HCl), nitric acid (HNO3), sodium chloride (NaCl), basic fuchsine (pararosaniline hydrochloride) and sodium metabisulfite were purchased from VWR International. Polyethylene glycol dithiol with a molecular weight of 1000 g.mol−1 (referred to as PEG double thiol) and periodic acid were purchased from Sigma-Aldrich (St. Louis, MO). Polyethylene glycol methyl ether thiol with a molecular weight of 2000 g.mol−1 (referred to as PEG 2000) was purchased from Laysan Bio (Arab, AL). Dichloromethane and methanol were purchased from Fisher Scientific, and mucins from bovine submaxillary gland were purchased from Cedarlane Laboratories (Burlington, ON). All the glassware used for the synthesis of AuNP was first thoroughly washed with aqua regia (3:1 HCl:HNO3) and rinsed with nanopure water.

Dimethylformamide (DMF, ≥99.9%) was purchased from VWR Chemicals. Chloroform-D (CDCl3, 99.8%) was purchased from Cambridge Isotope Laboratories Inc. Cellulose ester dialysis membranes (3.5-5 kDa MWCO) were purchased from Spectrum Laboratories Inc. All Plasma treatment was carried out using a Henniker Plasma Vacuum System HPT-200. pH measurements were taken using a Mettler Toledo, SG2 -SevenGo pH meter.
