Erythroid Cell Development Analysis

Bone marrow, spleen and fetal liver cells were stained for erythroid subsets using the antibodies listed below. Samples were analyzed by a FACSCantoII flow cytometer using FACSDiva software (Becton-Dickinson). Erythrocyte development in the bone marrow and spleen was analyzed as follows: Ter119medCD71high (proerythroblasts; R1), Ter119highCD71high (basophilic erythroblasts; R2), Ter119highCD71med (late basophilic and polychromatophilic erythroblasts; R3) and Ter119highCD71low (orthochromatophilic erythroblasts; R4). Cells were stained with the fluorescently labeled antibodies: Ter119-PE (eBioscience), CD71 Biotin (eBioscience), Streptavidin eflour 780 (eBioscience), CD49e PE (integrin α5), and CD49d FITC (integrin α4).

Samples were preincubated with 1 μg Fc-block (2.4G2 hybridoma; ATCC).
Cells were acquired either on the BD Biosciences LSR Fortessa or with a BD FACScan flow cytometer with DxP multi-color upgrades by Cytek Development Inc. (Woodland Park, NJ) and analyzed using FlowJo software (FlowJo LLC, Ashland, OR).
