Immunohistochemical Visualization of RFP-Positive Neurons

To analyze the distribution of the red fluorescent protein (RFP)-positive neurons, brain tissue sections were treated with 1% H2O2 (Sigma-Aldrich) for 15 min to quench endogenous peroxidases, incubated in the blocking buffer for at least 2 hours, and then incubated overnight in rabbit-anti-RFP antisera (1:2,000; Rockland Immunochemicals, Gilbertsville, PA) at room temperature. Sections were then rinsed in PBS, incubated in biotinylated donkey anti-rabbit IgG (1:500; Jackson ImmunoResearch, West Grove, PA) for 2 hours, incubated with peroxidase-conjugated avidin-biotin complex (1:200; ABC, Vector Laboratories, Burlingame, CA) for 1 hour, followed by the addition of 3, 3’-diaminobenzidine tetrahydrochloride, H2O2 and NiSO4 (DAB Peroxidase Substrate Kit, Vector Laboratories, Burlingame, CA) to visualize the reaction product.
