Protein Extraction and Western Blot Analysis

To obtain protein extracts, the cell pellets were lysed using M-PER mammalian protein extract reagent (78501, Thermo Fisher Scientific) containing protease (P8340, Sigma) / phosphatase (P5726, Sigma) inhibitor cocktail. Cell lysates were resolved on 4%-12% of Bis-Tris gels and transferred onto nitrocellulose membranes. After incubation with appropriate antibodies (Recombinant Anti-MMP14 antibody [Abcam, ab51074] or Beta-Actin Antibody [Santa Cruz, sc-1616]), blots were developed and visualized with Clarity ECL Western Blotting Substrates (Bio-Rad Laboratories Inc.).
