FU97 cell lysates were extracted using RIPA Lysis Buffer containing protease and phosphatase inhibitors (Beyotime), fractionated by SDS‐polyacrylamide gel electrophoresis, and electrotransferred to polyvinylidene difluoride (PVDF) membranes. After blocking in 5% skim milk in triethanolamine buffered saline with 1‰ Tween‐20 (TBST) for 1 h at room temperature, the membranes were incubated with primary antibodies (AFP [1:2500, Abcam, EPR20667], GPC3 [1:1000, HUABIO, EM1709‐60], ALB [1:500, HUABIO, ET1702‐55], E‐cadherin [1:10000, Abcam, EP700Y], N‐cadherin [1:5000, Abcam, EPR1791‐4], VIM [1:1000, Abcam, EPR3776], SNAIL [1:1000, Abcam, EPR21043], and GAPDH [1:5000, Abcam, EPR16891]) at 4°C overnight and then with peroxidase‐conjugated secondary antibodies (HRP Conjugated Goat Anti‐Rabbit IgG [1:5000, HUABIO]) for 1 h at room temperature. After washing thrice with TBST, the blot was soaked in Super ECL Detection Reagent (YEASEN) for 10 s. The membranes were then exposed to a film (Kodak) for 10 s in the dark.

DNA gel extraction kits (Omega Bio-tek, USA), AxyPrep plasmid miniprep kit (Axygen Scientific Inc, USA) were used for DNA fragments purification and plasmid extraction according to the manufacturer's instructions. The BCA protein quantification kit and T4 DNA ligase were from Vazyme Biotech (Nanjing, China). The following reagents were purchased from Sangon Biotech (Shanghai, China): urea, imidazole, Ni-NTA sefinose resin, dialysis membrane (cutoff MW, 14 KD), polyethylene glycol-20000 (PEG20000), 50 × TAE buffer, two-color pre-stained protein marker. The 2 × Hieff PCR Master mixture, nucleic acid gel stain (10000 × in water), and DNA marker were obtained from Yeasen Biotech (Shanghai, China). NcmECL Ultra Kit was obtained from NCM Biotech (Suzhou, China) and used for detection of Western blot.

Immunohistochemical Analysis of TSLP and Phospho-SYK in Human Skin Samples

Human skin tissue samples were obtained from PLA General Hospital with informed consent. A total of 10 samples were collected, and the related patient information is included in the Table S1 . Immunohistochemistry staining was performed on tissue slices 5 µm thick and fixed in paraffin. After antigen retrieval, tissue sections were stained with primary antibodies anti-TSLP (13778-1-AP, Proteintech, China) and anti-Phospho-SYK (Y348, #12293, SAB, USA) at 4 °C overnight. Slides were washed three times with TBST, and the secondary antibody HRP conjugated goat antirabbit IgG (HA1001, HuaBio, China) was incubated at 37 °C for 1 h. Following three washes with TBST, the slides were treated with a DBA substrate solution. Hematoxylin was used to counterstain the specimens.
