Immunofluorescence and Immunoblotting of ABCB4

Indirect immunofluorescence was performed on cells grown on glass coverslips after fixation with methanol at -20°C, as described (Delaunay et al., 2009) . In the case of HEK293 cells, coverslips were pre-coated with poly-L-lysine (Sigma-Aldrich, Saint-Quentin Fallavier, France). The monoclonal P3II-26 anti-ABCB4 was obtained from Enzo Life Sciences (Villeurbanne, France) and Alexa Fluor-labeled secondary antibodies were from Invitrogen-Life Technologies. Electrophoresis and immunoblotting were performed as described (Delaunay et al., 2009) using the monoclonal P3II-26 anti-ABCB4 antibody, and peroxidaseconjugated secondary antibodies from Rockland Immunochemicals (Gilbertsville, PA).
