CHO-K1 Cell Culture and Antibody Detection

Chinese hamster ovary cells (CHO-K1) were grown in 75-cm2 cell culture flasks (Corning) in Ham’s F-12 medium (BioWhittaker) supplemented with 10% fetal bovine serum (HyClone) and 1% penicillin-streptomycin (BioWhittaker). Trypsin-Versene (BioWhittaker) was used to detach adherent cells. Polyclonal anti-gp42 antibody serum (PB114) was used as previously described (27 (link)). Monoclonal antibody 3H3 (anti-gp42) was obtained as previously described (11 (link)). The HLA class II DQ monoclonal antibody (1a3) (catalog no. ab24265; Abcam) and anti-FLAG-M2 (catalog no. F1804; Sigma-Aldrich Chemical Company) were used to detect HLA class II and gp42, respectively. Monoclonal anti-FLAG M2 antibody (F1804) and polyclonal anti-FLAG antibody (F7425) were obtained from Sigma-Aldrich Chemical Company.
