Detecting AAV9-Mediated Gene Expression in Rat Tissues

Check if the same lab product or an alternative is used in the 5 most similar protocols
Two weeks after AAV9 vector transfer of the reporter gene (ZsGreen) into rat myocardium, 6 μm cryosections were prepared from rat heart, lung and liver tissues, and examined for green fluorescence protein expression under fluorescence microscopy (Olympus BX-51 microscope, Olympus, Melville, NY, USA). Transfected HEK293 cells, isolated adult rat cardiomyocytes exposed to H/R, and fresh frozen ventricular sections from study groups were fixed for 10 min in 4% formaldehyde (Beyotime Biotechnology, Shanghai, China), stained with primary antibodies targeting PIASy (sc-166706, sc-50348, Santa Cruz Biotechnology, Danvers, MA, USA) and Cav-3 (ab2912, Abcam, Cambridge, MA, USA), Nav1.5 (#ASC-005, Alomone Lab, Jerusalem, Israel), and SUMO2/3 (ab3754, Abcam, Cambridge, MA, USA), followed by incubation with goat anti-mouse IgG secondary antibody (#4408, Santa Cruz Biotechnology, Danvers, MA, USA) or goat anti-rabbit IgG secondary antibody (#8889, Cell Signaling Technology, Danvers, MA, USA). Nuclei were stained with 4′6-diamidino-2-phenylindole (DAPI, C1005, Beyotime Biotechnology) during secondary antibody incubations. Fresh ventricular tissues were paraffin-embedded, and 5 μm sections were stained with hematoxylin and eosin (H&E). An Olympus fluorescence microscope was used for imaging.

Western Blotting Protocol for Cardiac Tissue

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 2 h of reperfusion, rat ventricular tissues in the peri-infarct region were harvested, and the procedure for Western blotting was performed as described previously.[14 (link)] The primary antibodies used in this study were as follows: anti-Cx43 (1:1000, #3512S, Cell Signaling Technology), anti-phospho-Cx43-S368 (1:1000, #3511S, Cell Signaling Technology), anti-SUMO2/3 (1:500, ab3754, abcam, USA), anti-PIASy (1:1000, sc-166706, Santa Cruz Technology, Beverly, MA, USA), anti-voltage-gated Sodium Channel 1.5 (NaV1.5) (1:200, GTX54798, Gentex, San Antonio, TX, USA), and anti-plakophilin 2 (PKP2) (1:1000, GTX54579, Gentex). The secondary antibody (1:5000) was purchased from Cell Signaling Technology.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
