The doses of GRg3 (10, 20 and 40 mg/kg i.g) were selected according to its antidepressant-like effects15 (link). sertraline (15 mg/kg i.g) was prepared as a positive control in the behavioral assessments that based on the previous study24 (link).

Pharmacological Effects on Zebrafish Development

Adult wild type zebrafish were obtained from Carolina Biological Supply and have been housed at Brown University over several generations in multiple 10 and 20 gallon tanks. For breeding purposes, the zebrafish were kept in mixed male and female populations on a 14 hour light/10 hour dark cycle. The fish were fed a combination of Gemma fish food, frozen brine shrimp, and frozen bloodworms. Embryos were collected from the tanks at the beginning of the light cycle (“dawn”) and were immediately transferred to deep Petri dishes (Fisher 08-752-11Z) containing ‘egg water’ (0.06 g of Instant Ocean and 0.25 mg of methylene blue per liter of deionized water). The embryos were then treated with various dilutions of bupropion hydrochloride (Sigma B102), sertraline (Sigma S6319), or duloxetine hydrochloride (Sigma SML0474) and grown in an incubator at 28.5 °C with a 14 hour light/10 hour dark cycle at a density of 60–70 embryos per culture dish.

Chronic Stress and SSRI Treatment in Mice

The chronic stress model used consists in the aleatory, intermittent, and unpredictable exposure to different stressors during 5 weeks in C57BL/6J mice. Briefly, animals were randomly and alternately exposed to one of the following stressors for the time indicated for each assay: restraint in well-ventilated tubes for 6 h (20 (link)), tail suspension for 5 min (21 (link)), forced swimming for 5 min (22 (link)), cold temperature exposure (4°C) for 2 h (23 (link), 24 (link)), and 2 days of continuous overnight illumination (25 (link)).
To analyze the effect of SSRIs, mice were orally given 15 mg/kg/day of fluoxetine (Sigma-Aldrich) (26 (link)) or 20 mg/kg/day of sertraline (Sigma-Aldrich) (27 (link)), in a fresh solution prepared in the drinking water. The preparation of these solutions was performed taking into account the volume of water drunk daily by each mouse (5 ml) to reach the indicated dose.

Whole-Cell Recording of Kv1.5 Potassium Channels
