Optimizing Chromosome Doubling in Tammarin Rock

The application of Trifluralin was tested on the variety Tammarin Rock. This variety was screened in an earlier study, in which it exhibited good green plant regeneration but a relatively low frequency of chromosome doubling (30%) [11 (link)]. Eight donor plants were sown over two sowing dates (Table 5). Spikes 1–10 were harvested from the first sowing date, and spikes 11–20 were harvested from the second sowing date. Spikes were harvested over 6 days and stored at 4 °C for up to 2 days. The experimental design and protocol was the same as Experiment 1 with anthers from spikes 1–10 exposed to three treatments (control, 1 µM Trifluralin for 24 h and 1 µM Trifluralin for 48 h) and anthers from spikes 11–20 exposed to different treatments (control, 3 µM Trifluralin for 24 h and 3 µM Trifluralin for 48 h) (Table 7). As per Experiment 1, anthers were exposed to Trifluralin immediately following mannitol and n-butanol pretreatment at the start of the induction phase. Trifluralin (N13689, Sigma-Aldrich) was dissolved in sterile dimethyl sulfoxide (DMSO) (D2650, Sigma-Aldrich), and 10 or 30 µL of Trifluralin stock was added to 4 mL LIM to provide final concentrations of 1 or 3 µM, respectively.

Quantitative Determination of Pesticide Residues

Benoxacor (98.4% purity; InChIKey: PFJJMJDEVDLPNE-UHFFFAOYSA-N) and trifluralin (99.3% purity; InChIKey: ZSDSQXJSNMTJDA-UHFFFAOYSA-N; recovery standard) were purchased from Sigma-Aldrich (St Louis, MO, USA). The internal standard, acenaphthene (99.3% purity), was purchased from Accustandard (New Haven, CT, USA). The nonspecific inhibitor of cytochrome P450 enzymes, 1-aminobenzotriazole [20 (link)], was obtained from TCI (≥98% purity; InChIKey: JCXKHYLLVKZPKE-UHFFFAOYSA-N; Portland, OR, USA). Benzil, classical CES inhibitor [21 (link), 22 (link)], was purchased from ACROS Organics (≥99% purity; InChIKey: WURBFLDFSFBTLW-UHFFFAOYSA-N; New Jersey, NJ, USA). Ethacrynic acid, a frequently used inhibitor of GSTs [23 (link), 24 (link)], was provided by Enzo Life Sciences (≥98%; InChIKey: AVOLMBLBETYQHX-UHFFFAOYSA-N; Farmingdale, NY, USA). Pesticide grade ethyl acetate used for liquid-liquid extractions was supplied by Fisher Chemical (Fair Lawn, NJ, USA). Chemicals for the preparation of buffers were obtained from commercial sources and were of ACS grade (≥98% purity).

Adsorption of Organic Pollutants on Clays

High-performance liquid chromatography (HPLC)-grade methanol, acetonitrile and pH buffers (4.0, 7.0 and 10.0) were purchased from VWR (Atlanta, GA, USA). Analytical standards for lindane, diazinon, linuron, trifluralin, paraquat and ammonium acetate were purchased form Sigma Aldrich (St. Louis, MO, USA). Sulfuric acid (H2SO4, 95–98%), PCP and 2,4,6-TCP were purchased from Aldrich Chemical Co. (Milwaukee, WI, USA). Ethylene glycol and formic acid (HCOOH, 88%) were purchased from Thermo Fisher (Waltham, MA, USA). CM was obtained from BASF (Lampertheim, Germany) with a total surface area of approximately 850 m2/g, an external surface area of 70 m2/g and cation exchange capacity equal to 97 cmol/kg [28 (link)]. SM clay was obtained from the Source Clay Mineral Repository at the University of Missouri-Columbia with an estimated cation exchange capacity equal to 75 cmol/kg. The generic formula for these clays is: (Na,Ca)0.3(Al,Mg)2Si4O10(OH)2⋅nH2O. Clays were sieved through 45 μm to achieve uniform particle size [28 (link),29 (link)]. Ultrapure deionized water (18.2 MΩ) was generated in the lab using an Elga™ automated filtration system (Woodridge, IL, USA) and used in all experiments.

Trifluralin Evaluation on Leaves

Trifluralin, polyvinylpyrrolidone (PVP) K30 (molecule weight≈300g), sodium hydroxide (NaOH), ethanol, tri-Sodium citrate (Na3C6H5O7), silver nitrate (AgNO3) and ethylene glycol (EG) were purchased from Sigma-Aldrich (Ontario, Canada). Phosphate buffer saline (PBS) solution (0.1M, pH~7.4) was prepared by dissolving NaH2PO4 (0.2M) and Na2HPO4 (0.2M) in deionized water.
For sample preparation, 1mM of Trifluralin in mixed solution of acetone-PBS with volume ratio of 1:4 and 100μl of prepared solution was poured on the leaves surface and analyzed.

Pesticide Mixture Analysis Protocol

Pesticide mixtures, including triazines, organophosphorus pesticides (OPPs), and carbamates in acetonitrile (ACN), were purchased from AccuStandard (New Haven, CT, USA). Pure standards of chlorophenols, trifluralin, and methyl parathion were obtained from Sigma-Aldrich (Oakville, ON, Canada). Internal standards, including 3,5-dichlorophenol-d3, trifluralin-d14 and metolachlor-d6, were prepared from stock solutions sourced from CDN Isotopes (Pointe-Claire, QC, Canada). DVB particles (5 µm diameter) and high-density PLOT PDMS, used in the preparation of the membrane, were obtained from Supelco (Bellefonte, PA, U.S.A). Nanopure water was sourced from a Barnstead/Thermodyne Nanopure ultra-pure water system (Type 1 water grade) for method development. A mixture of standards at different concentrations was prepared in ACN by diluting stock solutions for preliminary experiments, method development, and preparation of calibration levels. Properties of the selected pesticides are provided in Table S1 of Supporting Information.
