Chemical Analysis Procedures Protocol

The N-hexane, ethyl acetate, methanol, ethanol, Folin–Ciocalteu reagent, gallic acid, Sodium carbonate, potassium hydroxide, diethyl ether, phenolphthalein, starch indicator were purchased from Sigma-Aldrich (Steinheim, Germany). Wijs solution (0.1 mol∙L1 iodine monochloride, acetic acid) was purchased from (Fluka (Steinheim, Germany)), chloroform, acetic acid by (lachner), potassium iodide, sodium thiosulfate by (CHEMPUR (Poland)). The methylene chloride, sodium chloride, sodium sulfate were provided by (Acros Organics (Great Britain)). All of the chemicals used for the analyses were of analytical grade, and all of the test solutions were freshly prepared prior to use.

Cytotoxicity and Apoptosis Evaluation

Fetal bovine serum (FBS) was purchased from Gibco (Carlsbad, CA, USA). Potassium iodide was purchased from Chempur (Piekary Slaskie, Poland). Acetic acid and dimethyl sulfoxide (DMSO) were purchased from POCH (Gliwice, Poland). Alexa Fluor 488 Annexin V/Dead Cell Apoptosis Kit was purchased from Life Technologies (Carlsbad, CA, USA). Caspase-Glo® 3/7 was purchased from Promega (Madison, WI, USA). JC-10 Mitochondrial Membrane Potential Assay Kit was purchased from Abcam (Cambridge, UK). RNA Extracol, NG dART RT kit, and SG qPCR Master Mix (2×) were obtained from EURx (Gdansk, Poland).

Chlorogenic acid, cyanidin, quercetin 3-glucoside, quercetin 3-rutinoside, and quercetin 3-rhamnoside were obtained from Extrasynthese (Lyon, France). NeoChlorogenic acid, cryptoChlorogenic acid, procyanidin C1 were purchased from PhytoLab (Vestenbergsgreuth, Germany). Ultrapurity water was prepared in the laboratory using a Simplicity Water Purification System (Millipore, Marlborough, MA, USA).

Procyanidin C1, procyanidin B1, procyanidin B2, neochlorogenic acid, cryptochlorogenic acid, and 3,5-dicaffeoylquinic acid were purchased from PhytoLab (Vestenbergsgreuth, Germany). Kaempferol was purchased from ICN Biomedicals (Costa Mesa, CA, USA). Ultrapurity water was prepared in the laboratory using a Simplicity Water Purification System (Millipore, Marlborough, MA, USA).
