Cytokine Quantification in Airway Inflammation

BAL fluids from the papain-induced airway inflammation model and supernatants acquired after in vitro CD4+ T cell activation and differentiation were stored at −80 °C. The quantities of cytokines were measured using the antibodies listed in Supplementary Table 3. Quantification of Amyloid-Beta Levels

To quantify levels of soluble and insoluble Aβ1–42, the well-established human Aβ42 ELISA kit (KHB3441, Invitrogen, CA, USA) was used. The samples were prepared according to the manufacturer’s protocol. Briefly, the cortex and the hippocampus from 11 month to 12-month old transgenic mice and their WT counterparts were removed from fresh brains and weighed. With the lysate buffer (5 mM guanidine HCl, 50 mM Tris-HCl, pH 8.0), samples were homogenized and centrifuged. Fractions were then analyzed according to the manufacturer’s protocol. Absorbance was determined for each well at 450 nm using a microplate reader (MPM6, Bio-Rad, Hercules, CA, USA).
