The WST-1 solution is a colorimetric assay reagent used for the quantitative determination of cell viability and proliferation. It contains the tetrazolium compound WST-1 (2-(4-Iodophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt), which is reduced by metabolically active cells to produce a water-soluble formazan dye. The intensity of the produced color is directly proportional to the number of viable cells, allowing for the assessment of cell proliferation and cytotoxicity.

Cell Viability Assay with 5-Aza

Cell viability was measured using the WST-1 assay. Briefly, cells were seeded in 96-well plates at 4,000 cells per well, and treated with different concentrations of 5-Aza in triplicate for an indicated period of time. At the end of the treatment, 20 μl of WST-1 solution (Beyotime Biotechnology, Shanghai, China) was added into each well and the cells were cultured at 37°C for an additional 2 h. Finally, the absorbance was measured at 450 nm and 690 nm using a microplate reader (Thermo Fisher Scientific, Vantaa, Finland). The data were calculated by optical density (OD) OD450-OD690 and represented the cell viability.

TRAIL-Induced Cell Viability Assay

Cell viability was assessed using the WST-1 assay. Cells were pretreated with different concentrations of CK for 24 h and then exposed to various concentrations of TRAIL for 24 h. The medium was then removed, and 10 μl per well of the WST-1 solution (Beyotime Biotechnology, Jiangsu, China) was added. After 4-h incubation, the absorbance was measured at 450 nm using a microplate reader (Bio-Rad, Hercules, CA, USA). All experiments were performed in triplicate and repeated at least three times. Cell viability is expressed as the percentage of the control, which was set to 100%.
