Western Blot Analysis of Phosphorylated MSK1

Twenty micrograms of protein was loaded for SDS-PAGE and transferred onto a PVDF membrane for Western blots. Western Blot Analysis of STAT1 and pSTAT1

MSCs were lysed in RNA immunoprecipitation (RIPA) buffer. Total protein concentrations were quantified using a BCA assay. 25μg of protein was loaded for SDS-PAGE and transferred onto a PVDF membrane for western blot. Primary antibodies for STAT1 (66545-1, Proteintech, Rosemont, IL) and pSTAT1 (MA5–15071, Thermo, Waltham, MA) were used at a 1:5000 and 1:1000 dilution, respectively, in 1X Tris Buffered Saline with 1% Casein (Bio-Rad, Hercules, CA). Secondary α-mouse HRP (A6154, Sigma-Aldrich, St. Louis, MO) or α-rabbit HRP (ab6721, Abcam, Cambridge, UK) were used at a 1:3000 dilution in 1X Tris Buffered Saline with 1% Casein. Membranes were exposed after addition of ECL (170–5060, Bio-Rad, Hercules, CA). Tubulin was detected with hFAB Rhodamine Anti-Tubulin Primary Antibody (12004166, Bio-Rad, Hercules, CA) at 1:3000 dilution.
