Berberine Hydrochloride Signaling Pathway

Signaling Pathway Modulation Assay

Phorbol 12-myristate 13-acetate (PMA) (#P8139), interleukin-1A (#I2778), oligomycin (#75351), antimycin A (#A8674), AICAR (#A9978) and forskolin (#F6886) were all purchased from Sigma–Aldrich. Human EGF (#8916), and human IGF-1 (#3093) were purchased from cell signaling technology (CST). PKC-specific inhibitor GO6983 was purchased from Selleckchem (#S2911). ON-TARGETplus SMARTpool human LRRK1 (#L-005320-00-0005), human LRRK2 (#L-049666-00-0005) and non-targeting control pool (#D-001810-10-20) siRNA were purchased from Horizon Discovery.

Elucidating Splicing Regulation by TCR Signaling

To elucidate the roles of signal transduction proteins to regulate splicing changes, each protein known to be regulated downstream of TCR signaling was targeted by at least two different inhibitors. Human primary CD4+ CD45RO- T cells were incubated with individual inhibitors for 1 hr and stimulated with bound anti-CD3 and soluble anti-CD28 antibodies for 48 hr. Cells were then harvested for RNA purification and low-cycle RT-PCR analysis for splicing quantifications. Optimizing Inhibitor Concentrations for Immune Cell Studies

MCC950 (Adipogen, San Diego, CA), glycine (Fisher Scientific, Waltham, MA) and potassium chloride (Fisher Scientific) were resuspended in deionized water. Go6983 (Selleck Chemicals, Houston, TX), Ac-Tyr-Val-Ala-Asp-chloromethylketone (Ac-YVAD-CMK or YVAD) (Cayman Chemical, Ann Arbor, MI), MI2 (Tocris Bioscience, Bristol, UK), PS1145 (Cayman Chemical), R406, and entospletinib (Selleck Chemicals), were all resuspended in DMSO. Monocytes were treated with the inhibitors or with an equivalent volume of the appropriate vehicle, for 40 min at 37°C and then infected or stimulated as described above. MCC950, Go6983, MI2, PS1145, R406 and entospletinib were all added to infected cells in half log titrations to determine the concentrations of the inhibitors that did not induce cell death or reduce infection efficiency.

Inhibition of PKC Signaling Pathway

Immortalized Mouse Podocyte Culture

Synthesis and Characterization of Ricefield Eel Peptide
