Capturing and Collaring Asian Black Bears

Asian black bears were captured from May to June 2018 using barrel traps baited with honey. A total of five traps were set within 5 km of each other, and the trapped bears were immobilized with tiletamine hydrochloride and zolazepam hydrochloride (Virbac, Carros, France, 8 mg/1kg of body weight). Basic body measurements were taken, and premolar extraction was performed for age determination. Bears over 40 kg were equipped with microchips for individual identification and GPS/camera collars (GPS Vertex Lite Collars; Vectronic Aerospace GmbH, Berlin, Germany). The collars (1.2 kg) weight less than 3% of body mass of bears. Bear capture and handling were approved by the Institutional Animal Care and Use Committee of Tokyo University of Agriculture and Technology. All experimental procedures used in the bear research followed the Guidelines Concerning Animal Experimentation of the Tokyo University of Agriculture and Technology and the Mammal Society of Japan (Mammal Society of Japan 2020).

Microwave Ablation Effects on Tibial Bone in Sheep

For the experiment permission was obtained from the medical ethical committee of the HongHui hospital, Xi’an, China. Ablations were performed in tibia of male sheep (Small-tail Han) aged 1–1.5 years with aimed mass of 50 kg (in vivo experiment: range 45–50 kg). Three settings were examined for both a one week- and a six weeks follow-up group: 60W 10min, 60W – 20 min and 80W – 10 min. For each group four sheep were included, leading to a total of 24 sheep. As controls the contralateral tibias were used. Sheep were randomly divided in the one- or six week group. Before starting the procedure (Fig 1A), the sheep were sedated with an intramuscular injection of 5 ml Shutai (1: 1 combination of Tiletamine 50mg/ml & Zolazepam hydrochloride 50mg/ml) (Virbac, France) and subsequently intubated (Fig 1B. During the complete procedure inhaled isoflurane (1–5%) was used for general anesthesia and a maintenance dose up to 2.5ml Shutai was given in case deemed necessary. For post-procedural analgesia meloxicam injections were used. After the follow-up period sheep were euthanized by an experienced veterinarian (bleed out under intramuscular sedation). Sheep with a (deep) infection or fracture were excluded from mechanical analysis to prevent finding differences in strength based on other factors than the mechanical effect of MWA on bone.

Evaluation of IBDV Vaccines in China

The nVarIBDV FJ2019-01 strain (GenBank: MZ736578 or MZ044944) was locally available in our laboratory (Hou et al., 2022 (link)). The live viral vector vaccine (VAXXITEK, recombinant vaccine generated by inserting the IBDV VP2 gene into the HVT genome; Boehringer Ingelheim, China), the IBDV immune complex vaccine (TRANSMUNE, W2512 G-61 strain, CEVA, China), and VP2 subunit vaccine (YEBIO, Qingdao, China), the attenuated live vaccine (B87 strain, Liaoning Yikang, Liaoyang, China), and the attenuated live vaccine (W2512 G-61 strain; CEVA, China) were commercially licensed by the Chinese Ministry of Agriculture and used to against vvIBDV and/or classical IBDV in China.
The SPF chickens (White Leghorn) were purchased from Jinan SPAFAS poultry co., LTD (Shandong, China). The broilers were purchased from Fujian Sunner Development Co., LTD (Fujian, China). The FAAS's Institutional Animal Care and Use Committee guided all the animal experiments, performed following the animal ethics guidelines and approved protocols. Furthermore, all husbandry procedures followed the Animal Welfare Act and the Guide for the Care and Use of Laboratory Animals. All chickens were euthanized with Tiletamine Hydrochloride and Zolazepam Hydrochloride injections (Virbac, Carros, France), minimizing suffering.

Forced Expiratory Lung Function Measurement

Twenty-four hours after the last challenge, mice were anesthetized with an anesthetic solution containing tiletamine hydrochloride and zolazepam hydrochloride (25 mg/kg, Virbac S. A., France) and xylazine hydrochloride (10 mg/kg, Chang Sha Best Biological Technology Institute Co., Ltd, Hunan, China) via intraperitoneal injection. Mice were tracheostomized, cannulated endotracheally, and ventilated with a pneumotachograph connected to a transducer (EMMS, Hants, UK) at 250 breaths/min and tidal volume of 250 ml. To mimic clinical spirometry, the lungs of mice were inflated to a set tracheal pressure, and then exposed to a large negative pressure reservoir, forcing the mouse to exhale as quickly as possible. Variables, including the FVC, FEV50, MMEF, FEF50 and FEF75, were calculated.

We then fit each bear with microchips and GPS collars mounted with digital cameras (GPS Vertex Lite Collars; Vectronic Aerospace GmbH, Berlin, Germany, 29.97 fps, pixel size: 1,920 × 9.97, dimensions: 70 × 70 × 84 mm, 1,200 g). Due to the weight of the GPS collars, we only included bears weighing >40 kg in this study. We dropped the collars using a remote control at the end of the programmed lives of the cameras. We performed all our bear capture and handling in accordance with the guidelines for animal research established by the Mammal Society of Japan (2009) .
