Analyzing AMPK Regulation via Western Blotting

Cells were lysed with RIPA buffer (Sigma) containing 1 mM PMSF
(Biyuntian) and proteinase inhibitor cocktail (Sigma). The following antibodies
were used: anti-ATP5A (clone 15H4C4, Abcam), anti-ATPB (clone 3D5, Abcam), AMPK
(Cell Signaling), AMPK Pi (Thr172, clone 40H9, Cell Signaling). ECL substrate
(Biorad) was used to detect proteins in Western blotting and the blot bands were
quantified by using Image J.
